Enhanced biosynthesis of D-tagatose from maltodextrin through modular pathway engineering of recombinant Escherichia coli

Summary: In this study, the biosynthesis efficiency of D-tagatose was enhanced by optimizing vectors and improving the expression of rate-limiting enzymes. The accumulation of intermediates was further increased by deleting genes in the bypass pathways. This work demonstrates the use of modular engineering to enhance D-tagatose production using whole-cell catalysts.

Metabolic Engineering of Escherichia coli for Hyperoside Biosynthesis

Summary: Hyperoside, a compound with higher bioactivities than quercetin, was efficiently synthesized from quercetin by co-expressing UGT from Petunia hybrida and GalE in E. coli. Additional enzymes Pgm and GalU were introduced to enhance UDP-glucose synthesis, leading to improved hyperoside production. Through fermentation optimization, hyperoside production was increased, and substrate-fed batch fermentation further increased production to 831.6 mg/L with a high conversion ratio and specific productivity. This efficient pathway can be applied for glycosylation of other flavonoids and bioactive substances.

This study demonstrated the use of six industry-relevant animal and plant by-products as complementary substrates in functional screens, suggesting temperature as a screening parameter to potentially discover new broad-substrate range and robust proteases.

Summary: This study demonstrates the impact of cellular manganese/zinc (Mn/Zn) ratios on bacterial capsular polysaccharide (CPS) production, showing that higher Mn/Zn ratios increase CPS production via the Mn-dependent activation of the phosphoglucomutase Pgm. These findings provide a direct role for Mn/Zn homeostasis in regulating CPS expression levels and further support the ability of metal cations to act as important cellular signaling mediators in bacteria.

Summary: The genome of sponge-associated microorganism Pseudoalteromonas sp. PA2MD11 contains a variety of carbohydrate-active enzymes with potential for degrading marine polysaccharides, including GHs and PLs families. The complete metabolism of agar and alginate by PA2MD11 was predicted, and structural protein analyses showed conservation of catalytic machinery and suitable industrially-relevant features in the enzymes. This bacterium is a potential source of biocatalysts for marine polysaccharide degradation and production of pharmacologically active oligosaccharides.

Comparison of Hyaluronic Acid Biosynthetic Genes From Different Strains of Pasteurella multocida
