A data compendium associating the genomes of 12,289 Mycobacterium tuberculosis isolates with quantitative resistance phenotypes to 13 antibiotics

[Anonymous]

Summary: This study presents a comprehensive resistance prediction for tuberculosis using a large dataset of Mycobacterium tuberculosis isolates. The data includes whole-genome sequencing and minimum inhibitory concentrations to 13 antitubercular drugs. The dataset provides valuable information on the genotypic and phenotypic characteristics of drug resistance, and has the potential to advance our understanding of rare resistance phenotypes. The open-source nature of the data compendium encourages future research in the field.

Genomic signatures of pre-resistance in Mycobacterium tuberculosis

Arturo Torres Ortiz et al.

Summary: Recent advances in bacterial whole-genome sequencing have led to a comprehensive catalog of antibiotic resistance genomic signatures in Mycobacterium tuberculosis. The study found that certain genotypes in M. tuberculosis are more prone to acquiring resistance, with specific loci and genomic polymorphisms associated with a higher risk of resistance acquisition. By identifying markers of future antibiotic resistance, targeted therapy could be implemented to prevent resistance emergence in M. tuberculosis and other pathogens.

Prisons as ecological drivers of fitness-compensated multidrug-resistant Mycobacteritum tuberculosis

Sebastian M. Gygli et al.

Summary: In Georgia, 63% of MDR-TB cases were due to patient-to-patient transmission, with compensatory mutations and patient incarceration playing independent roles in transmission. Prisons fuel the epidemic of MDR-TB in Georgia by acting as ecological drivers of fitness-compensated strains with high transmission potential.

Summary: The study aimed to investigate the minimum level of heteroresistance predicting poor tuberculosis treatment outcomes. Retrospective analysis of isolates from 61 patients showed that heteroresistance <1% is not associated with poor treatment outcomes.

Drug resistance, fitness and compensatory mutations in Mycobacterium tuberculosis

Amel Kevin Alame Emane et al.

Summary: In order to eradicate tuberculosis, it is crucial to reduce the transmission of Multi-Drug-Resistant and eXtensively Drug Resistant strains. The fitness costs of different resistance mutations and the compensatory mutations largely determine the frequency of certain mutations in highly transmitted clusters. The compensatory mutations illustrate how bacteria adapt to overcome the effects of antibiotics, providing insights into the evolution of mycobacteria under stress.

Quantifying transmission fitness costs of multi-drug resistant tuberculosis

Summary: The study introduces a method for phylodynamic analysis of Mycobacterium tuberculosis, showing that drug-resistant strains incur a transmission fitness cost. The estimates are robust to different prior distributions on resistance acquisition rate, but are affected by dataset size.
