A tool for translating polygenic scores onto the absolute scale using summary statistics

Oliver Pain et al.

Summary: There is a growing interest in the clinical use of polygenic scores, and this study presents a method for converting polygenic scores to the absolute scale for binary and normally distributed phenotypes. The method uses summary statistics and has been validated using data from the UK Biobank sample. This study provides a useful tool for accurately interpreting polygenic scores using only summary statistics.

Sample sizes for saturation in qualitative research: A systematic review of empirical tests

Monique Hennink et al.

Summary: This review explores empirical studies on saturation in qualitative research to determine sample sizes and assessment strategies. The findings suggest that commonly used qualitative research methods typically require a small number of interviews or focus group discussions to reach saturation, although these results may only apply to studies with homogenous populations and clearly defined objectives.

Summary: The study found that the majority of individuals undergo genetic testing out of curiosity, but only a small number correctly understood and interpreted polygenic risk scores. Over half of the participants experienced negative psychological reactions after receiving their results.

Development of a clinical polygenic risk score assay and reporting workflow

Limin Hao et al.

Summary: The first report from the GenoVA Study provides insights into the development of a clinical polygenic risk score assay and discusses the challenges of implementing and reporting the results. The study developed a clinical genotype array-based assay for multiple conditions and analyzed prospective samples to determine the frequency of high-risk polygenic risk scores. The research highlights the importance of developing resources and workflows for using polygenic risk score information in patient care.

Natalie T. Boutin et al.

Summary: The Mass General Brigham Biobank is a large repository of biospecimens and data aimed at supporting translational research on disease phenotypes. It has enrolled over 135,000 participants, generated genomic data on more than 65,000 participants, and served numerous institutional studies. However, it faces challenges in terms of funding sustainability, integration into clinical workflows, and promoting diversity and equity in research resources.

Summary: Polygenic risk scores (PRSs) aggregate multiple small effects of alleles in the human genome to estimate an individual's risk of disease or disease-related traits. By addressing key issues, PRSs can be used responsibly to improve human health.

The limits of personalization in precision medicine: Polygenic risk scores and racial categorization in a precision breast cancer screening trial

Jennifer Elyse James et al.

Summary: Population-based genomic screening is a new approach to disease prevention, but the lack of diversity and debates on the use of racial and ethnic categories in genomics raise questions on translating genomic knowledge into clinical practice. This study explores challenges in using race or ethnicity as a risk factor in precision breast cancer risk assessment, shedding light on the deployment of these categories in producing genomic knowledge and medical practice. The conflation of social and biological categories of difference can impact risk prediction, highlighting potential harms of using under-theorized and ambiguous categories in genomic medicine.

Summary: The survey study revealed that practitioners tend to overestimate the probability of disease both before and after testing for common diseases. Overestimation of pretest probability was observed in all scenarios, while adjustments in probability after positive or negative results varied by test types.

Summary: The study found that while genome sequencing did not significantly increase the percentage of participants changing health behaviors post-disclosure, high-risk polygenic predictions were associated with increased odds of behavior change. Additionally, genome sequencing did not distress participants and may indirectly impact health behaviors through positive psychological responses.

Polygenic risk scores in the clinic: Translating risk into action COMMENT

Anna C. F. Lewis et al.

Summary: PRS is recognized as a useful tool for risk stratification and personalized preventive care, but its clinical utility depends on contextualizing risk information and translating it into action. Best practices may involve integrating PRS into an absolute risk model and using guideline-driven care linked to specific risk thresholds. Since this approach is not widely available for most diseases, alternative methods for presenting risk and linking it to appropriate clinical action may be necessary. Transparent communication of the imprecision in both risk estimates and action thresholds for PRS is essential for providers and patients.
