Biodistribution and Dosimetry Evaluation for a Novel Tau Tracer [18F]-S16 in Healthy Volunteers and Its Application in Assessment of Tau Pathology in Alzheimer's Disease

Summary: This study reported a fully automated radiosynthetic procedure of a novel tau tracer [F-18]-S16 and evaluated its safety, biodistribution, and dosimetry in healthy volunteers as well as its potential utility in Alzheimer's disease (AD). The results showed that [F-18]-S16 could be synthesized automatically with favorable biodistribution and safety in humans. [F-18]-S16 PET demonstrated high image quality for imaging tau deposition in AD and distinguishing AD from healthy controls.

Functional Abnormality Associated With Tau Deposition in Alzheimer's Disease - A Hybrid Positron Emission Tomography/MRI Study

Summary: This study investigated tau deposition and its impact on functional connectivity in Alzheimer's disease through PET/MRI scans and neuropsychological assessments. Higher tau accumulation was found in specific brain regions of AD patients compared to healthy controls, leading to decreased functional connectivity. Patients with positive functional connectivity were older, had better cognitive performance, and lower tau accumulation compared to patients with negative functional connectivity. The combination of tau-PET and rs-fMRI could potentially help predict AD progression.
