Lost and found: Frogs in a biodiversity hotspot rediscovered with environmental DNA

Carla Martins Lopes et al.

Summary: The study successfully used environmental DNA (eDNA) metabarcoding to detect DNA traces of 30 target amphibian species in Brazil, including four declined species, two locally disappeared species, and one species not seen since 1968. This highlights the potential of eDNA in biodiversity monitoring and conservation biology for evaluating the persistence and distribution of threatened species.

Estimating fish population abundance by integrating quantitative data on environmental DNA and hydrodynamic modelling

Keiichi Fukaya et al.

Summary: The study demonstrates that environmental DNA (eDNA) analysis can be a powerful tool to estimate population abundance of aquatic macroorganisms. By integrating knowledge of the processes governing eDNA distribution with numerical hydrodynamic models, a Bayesian inference of a generalized linear model can be used to estimate population abundance. The method was shown to provide comparable population abundance estimates to traditional methods and can also identify sources of exogenous eDNA input.

Universal performance of benzalkonium chloride for the preservation of environmental DNA in seawater samples

Toshiaki Jo et al.

Summary: This study examined the performance of benzalkonium chloride (BAC) in preserving eDNA in seawater samples, finding that BAC treatment can effectively suppress the degradation of eDNA and increase the eDNA yields. Additionally, the use of BAC can preserve both the quantitative and qualitative information of different types of eDNA in water, reducing false-negative detection of eDNA.

Integrating physiology and environmental dynamics to operationalize environmental DNA (eDNA) as a means to monitor freshwater macro-organism abundance

Matthew C. Yates et al.

Summary: Research has shown strong correlations between eDNA concentrations and organism abundance in laboratory experiments, but weaker correlations in natural environments due to various influencing factors. Future research must address these factors, including integrating organism physiology, eDNA dynamics, and key environmental parameters to improve accuracy. Despite challenges, recent developments indicate potential for eDNA to provide rough quantitative estimates of abundance in natural ecosystems when models incorporate physiology and eDNA dynamics.

Complex interactions between environmental DNA (eDNA) state and water chemistries on eDNA persistence suggested by meta-analyses

Toshiaki Jo et al.

Summary: This study analyzed 26 papers on the estimation of first-order eDNA decay rate constants, revealing the complex interactions between environmental factors and eDNA states that influence eDNA persistence. It highlights the importance of considering environmental conditions, as well as cellular and molecular states, to fully understand the mechanism of eDNA persistence and degradation.

Water pre-filtration methods to improve environmental DNA detection by real-time PCR and metabarcoding

Kazuto Takasaki et al.

Summary: The study introduced a pre-filtration method to improve the detection of fish species in environmental samples. The pre-filtration did not impact the number or identities of fish species detected, but significantly reduced variance among replicate samples through pore sizes. Additionally, PCR amplification was enhanced in environmental samples containing PCR inhibitors.

Trade-offs between reducing complex terminology and producing accurate interpretations from environmental DNA: Comment on Environmental DNA: What's behind the term? by Pawlowski et al., (2020)

Naiara Rodriguez-Ezpeleta et al.

Summary: The term eDNA should be used to refer to the pool of DNA isolated from environmental samples. The proposed two-level terminology specifying sampling environment and targeted taxa might hinder clear communication about environmental DNA and its use in biomonitoring. It overlooks the fundamental distinction of the type of DNA from which ecological interpretations are derived.

Jake J. Ponce et al.

Summary: The study demonstrates that on-site eDNA technology is effective for detecting and quantifying New Zealand mud snails, increasing positive detection rates and showing a positive correlation between mud snail eDNA concentration and individual numbers.

The Application of PMA (Propidium Monoazide) to Different Target Sequence Lengths of Zebrafish eDNA: A New Approach Aimed Toward Improving Environmental DNA Ecology and Biological Surveillance

Takaya Hirohara et al.

Summary: Environmental DNA (eDNA) analysis provides more sensitive and efficient biological monitoring compared to traditional methods, but interpretation of results may be prone to Type I errors due to potential spatial or temporal gaps between target eDNA and sampled area sources. Additionally, eDNA surveillance cannot distinguish between living and non-living sources, making it challenging to control for errors. By applying PMA to eDNA analysis, researchers can differentiate intact cells from disrupted cells, providing new insights into the degradation processes of total eDNA and intracellular eDNA for more reliable conservation efforts.
