It identifies different subtypes of PFC projection neurons and uncovers topographic rules and target selection patterns. Distinct thalamocortical circuits underlie allodynia induced by tissue injury and by depression-like states

Summary: In mice, tissue injury and depression can both cause pain hypersensitivity through distinct thalamocortical circuits involving glutamatergic neurons. While the posterior thalamic nucleus to primary somatosensory cortex circuit mediates allodynia from tissue injury, the parafascicular thalamic nucleus to anterior cingulate cortex circuit mediates allodynia associated with depression-like states. Manipulating these circuits affects allodynia from either tissue injury or depression-like states separately.

Disrupted population coding in the prefrontal cortex underlies pain aversion

Summary: This study used in vivo endoscopic calcium imaging to investigate the effects of inflammatory pain on the functional connectivity of PFC neurons. Inflammatory pain was found to disrupt this connectivity and reduce nociceptive response, which was restored by ketamine to produce anti-aversive effects. The results suggest that population activity in the PFC critically regulates pain and could be an important therapeutic target.
