Heterologous expression of a cryptic gene cluster from a marine proteobacterium Thalassomonas actiniarum affords new lanthipeptides thalassomonasins A and B

Summary: This study successfully produced new lanthipeptides through heterologous expression using a cryptic biosynthetic gene cluster (BGC) of the marine proteobacterium Thalassomonas actiniarum. This is the first report of heterologous production of lanthipeptides derived from proteobacterial origin, which may lead to the production of new lanthipeptides by utilizing similar BGCs.

Discovery, Structure Correction, and Biosynthesis of Actinopyrones, Cytotoxic Polyketides from the Deep-Sea Hydrothermal-Vent-Derived Streptomyces sp. SCSIO ZS0520

Summary: In this study, three new actinopyrone derivatives were isolated from a deep-sea hydrothermal vent bacterium. The structures and absolute configurations of these compounds were determined using various analytical techniques. One of the compounds exhibited notable cytotoxicity against human cancer cells, suggesting its potential as an antitumor agent.

Total synthesis of himastatin

Summary: The study presents a concise total synthesis method for himastatin, a natural product with an unusual homodimeric structure. The final-stage dimerization strategy used in the synthesis was inspired by a detailed understanding of the compound's biogenesis. By combining this approach with a modular synthesis, a series of designed derivatives of himastatin, including synthetic probes to investigate its antibiotic activity, were efficiently accessed.

Biosynthetic potential of the global ocean microbiome

Summary: Natural microbial communities are diverse and offer great potential for the discovery of enzymes and biochemical compounds. However, studying this diversity and assigning the synthesis of compounds to their hosts is challenging. In this study, we integrated microbial genomes from various sources and discovered thousands of new biosynthetic gene clusters, including in previously unsuspected phylogenetic groups. We identified a lineage rich in biosynthetic gene clusters and characterized the structures and enzymology of bioactive compounds. This research demonstrates the value of microbiomics-driven strategies in exploring previously undescribed enzymes and natural products.

Identification and Heterologous Expression of the Kendomycin B Biosynthetic Gene Cluster from Verrucosispora sp. SCSIO 07399

Summary: SCSIO07399 strain of Verrucosispora sp. produces ansamycin-like polyketides kendomycin B-D with potent antibacterial activities and moderate tumor cytotoxicity. The biosynthetic gene cluster for kendomycin B was identified, and the essential role of the kmy cluster in biosynthesis was confirmed through gene disruption and heterologous expression. Insights into kendomycin B biosynthesis were gained and a foundation was set for further research into exploiting the carbacylic ansa scaffold.
