Strategies of pluripotent stem cell-based therapy for retinal degeneration: update and challenges

Summary: Stem cell-based therapy for retinal degeneration is progressing from research to clinical stage globally. The safety of induced pluripotent stem cell (iPSC)-derived retinal pigment epithelium (RPE) transplantation has been confirmed, and clinical studies on transplantation of iPSC-derived retina for treating retinitis pigmentosa (RP) has begun. Collaboration between ophthalmologists and basic researchers is expected to enable the practical application of stem cell-based therapy for retinal degeneration.

Functional assessment of cryopreserved clinical grade hESC-RPE cells as a qualified cell source for stem cell therapy of retinal degenerative diseases

Summary: The study found that cryopreserved hESC-RPE cells retained specific gene expression, morphology, and functions similar to induced RPE cells, including phagocytic capability. However, the secretion of PEDF was significantly reduced in cryopreserved hESC-RPE cells. In rat experiments, there was an initial decrease in b-wave amplitude at 4 weeks post transplantation with cryopreserved hESC-RPE cells compared to induced cells, but no significant differences were observed at 8 and 12 weeks. This indicates that cryopreserved hESC-RPE cells are still suitable for cell-based therapy in retinal degenerative diseases even after cryopreservation and thawing.

Xeno-free cryopreservation of adherent retinal pigmented epithelium yields viable and functional cells in vitro and in vivo

Thawed cells exhibit high viability, morphology, pigmentation, and gene expression characteristic of mature RPE cells, and demonstrate stability under long-term storage. Implanted cells in a mammalian model showed survival, increased maturation marker expression, and functional attributes similar to controls, indicating potential for treating diseases like AMD.
