In silicodesigning of multi-epitope vaccine construct against human coronavirus infections

Arpita Devi et al.

Summary: Single-stranded RNA viruses have long been known to cause various diseases, with the recent pandemic of the novel coronavirus SARS-CoV-2 increasing their significance. Research is ongoing to develop a vaccine against SARS-CoV-2, with a focus on designing a multi-epitope vaccine construct using epitopes from the spike protein of human CoVs. Immuno-informatics techniques have been used to predict and design B- and T-cell epitopes on the spike proteins for the vaccine construct, which has been shown to have antigenic and non-allergenic properties.

Summary: The study identified outer membrane proteins as vaccine candidates against Escherichia albertii using a reverse vaccinology approach. A potential vaccine model was designed with the Fimbrial usher protein as the candidate, resulting in highly immunogenic V6 construct. This construct has the potential for pan-therapy against different strains of E. albertii and requires further in vitro and in vivo testing.

Immunoinformatics Design of Multi-Epitope Peptide-Based Vaccine Against Schistosoma mansoni Using Transmembrane Proteins as a Target

Rodrigo C. O. Sanches et al.

Summary: In this study, a robust immunoinformatics approach was applied to construct a chimeric antigen targeting S. mansoni transmembrane proteins, resulting in a stable and non-allergenic multi-epitope vaccine with the potential to induce B-cell and IFN-gamma based immunity. The molecular docking and dynamics showed stable interactions with the immune receptor TLR 4, indicating that the multi-epitope vaccine might stimulate humoral and cellular immune responses as a potential candidate against schistosomiasis.

Summary: The study aims to design a vaccine against Klebsiella aerogenes by combining immunological and molecular docking approaches. Researchers synthesized B-cell, T-cell, and IFN-gamma epitopes of three highly antigenic proteins and utilized docking techniques to confirm the compatibility of the vaccine with receptors. Further studies are essential to confirm the safety features and immunogenicity of the vaccine.

Pan-vaccinomics approach towards a universal vaccine candidate against WHO priority pathogens to address growing global antibiotic resistance

Saba Ismail et al.

Summary: The study identified a potential universal vaccine candidate against 18 multi-drug resistance bacterial pathogens using in-silico approaches. Nine B-cell-derived T-cell antigen epitopes with affinity to the dominant HLA allele (DRB1*0101) were screened, and a multi-epitope peptide vaccine construct (MEPVC) with beta-defensin adjuvant was designed. Molecular docking and MD simulation were conducted to investigate the binding affinity and molecular interaction of MEPVC with human immune receptors. The in-silico construct is expected to be useful in assessing its immune protection effectiveness against rising antibiotic resistance worldwide.

Summary: In this study, a multi-epitope subunit vaccine for Kyasanur forest disease virus (KFDV) was designed using in-silico methods. The vaccine candidates showed potential cross-protection against Alkhurma hemorrhagic fever virus and strong affinity with toll-like receptor-2 (TLR-2) was observed. Various tools were utilized to ensure the stability and efficiency of the vaccine constructs, paving the way for future vaccine and diagnostic kit development.
