Detailed Electronic Structure, Physico-Chemical Properties, Excited State Properties, Virtual Bioactivity Screening and SERS Analysis of Three Guanine Based Antiviral Drugs Valacyclovir HCl Hydrate, Acyclovir and Ganciclovir

Summary: This study investigates the structure and properties of three commonly used antiviral drugs, Valacyclovir HCl hydrate, acyclovir, and ganciclovir. The electronic structure and molecular mechanics methods were employed to analyze the molecular properties and bioactivity prediction. The results demonstrate the potential of these drugs as photosensitizers with excellent light-harvesting efficiency. Additionally, the adsorption of these drugs on a graphene quantum dot surface enhances their Raman activity, suggesting the use of Surface Enhanced Raman Spectra (SERS) for drug detection. Interestingly, the assembly of drug molecules on graphene shows enhanced biological activity compared to individual drug molecules.

Summary: New nano complexes of Co(II), Ni(II), Cu(II), Zn(II), Fe(III), and oxovanadium(IV), dioxouranium(VI) with a pyridazinone-acid hydrazone ligand, DCNHP (H2L), were synthesized and characterized. The complexes exhibited different geometrical structures and showed thermal stability. The antimicrobial activity study revealed enhanced activity upon complexation, and the antitumor screening indicated inhibition of Hepatocellular carcinoma cell line (HepG-2) viability. Molecular modeling and docking studies indicated strong interactions with the VEGFR-2 enzyme, similar to a known HCC inhibitor, sorafenib. The electrical conductivity study showed that the nano Cu(II) complex displayed higher values than the free ligand, acting as a semiconductor.

Summary: Novel nano-sized metal complexes derived from ACV were synthesized and characterized, showing good thermal stability and nano-scale size. Enhanced antimicrobial and antitumor activities were observed upon complex formation, with the Cu(II) complex potentially inducing cancer cell death through apoptosis/necrosis pathways. The Cu(II) complex also exhibited stronger binding affinity to DNA compared to the free ligand. The electrical conductivity measurements indicated that the complexes behave as semiconductors.
