Yields of mesenchymal stromal cells from synovial fluid reflect those from synovium in patients with rheumatoid arthritis

Yuji Kohno et al.

Summary: This study aimed to determine whether the yield of synovial fluid MSCs could predict the ultimate yield of primary MSCs from the synovium of patients with RA. The results showed a positive correlation between the number of synovial fluid cells and synovial cells, and the synovial fluid MSCs exhibited MSC-like characteristics and multi-differentiation potential. These findings reveal the feasibility of using synovial fluid MSC yield to predict the synovial MSC yield.

2021 American College of Rheumatology Guideline for the Treatment of Juvenile Idiopathic Arthritis: Recommendations for Nonpharmacologic Therapies, Medication Monitoring, Immunizations, and Imaging

Karen B. Onel et al.

Summary: This study provides recommendations for the management of juvenile idiopathic arthritis, focusing on nonpharmacologic therapies, medication monitoring, immunizations, and imaging. The guideline emphasizes the importance of improving patient quality of life and involving patients and caregivers in decision-making. However, the evidence quality for all recommendations is low.

Summary: This article provides updated guidelines for the pharmacologic management of juvenile idiopathic arthritis (JIA), focusing on different forms of JIA and providing recommendations for treatment and tapering of medications. The evidence for these recommendations is generally low quality, but the inclusion of patients and caregivers in the decision-making process strengthens the relevance and applicability of the guideline.

A Comparison of International League of Associations for Rheumatology and Pediatric Rheumatology International Trials Organization Classification Systems for Juvenile Idiopathic Arthritis Among Children in a Canadian Arthritis Cohort

Jennifer J. Y. Lee et al.

Summary: This study compared the ILAR and PRINTO JIA classification criteria in a cohort of JIA patients. The results showed that a large proportion of JIA patients were unclassifiable using the PRINTO criteria, and the alignment with clinicobiologic subtypes and adult forms of arthritis was not as good as the ILAR criteria.

p21-/- Mice Exhibit Spontaneous Articular Cartilage Regeneration Post-Injury

Christina L. Jablonski et al.

Summary: The absence of p21 in mice resulted in enhanced articular cartilage regeneration post-FTCD, with increased cartilage thickness at 1 and 2 weeks post-FTCD. This study suggests that p21 plays a crucial role in endogenous articular cartilage regeneration.

Summary: This meta-analysis study found that intra-articular mesenchymal stromal cell injections were not superior to placebo in pain relief and functional improvement for patients with knee osteoarthritis. However, compared with hyaluronic acid, MSC injection showed better pain relief and functional improvement.

Multi-pronged approach to human mesenchymal stromal cells senescence quantification with a focus on label-free methods

Weichao Zhai et al.

Summary: The study investigates alternative methods for quantifying the senescence of hMSCs, aiming to increase the final output of therapeutic-grade hMSC cultures. The advanced flow cytometry and autofluorescence methods show statistically significant values and good fold difference ranges between early and senescent passage hMSCs, providing a label-free approach for evaluating cellular senescence in hMSCs.

Cell size is a determinant of stem cell potential during aging

Jette Lengefeld et al.

Summary: The size of stem cells plays a crucial role in their function, with small size being important for maintaining stem cell potential. Enlargement of hematopoietic stem cells under conditions of decreased function is associated with reduced proliferation and altered metabolism. Preventing stem cell enlargement can help prevent the loss of stem cell potential in exhausting conditions.

Uncharted waters: mesenchymal stem cell treatment for pediatric refractory rheumatic diseases; a single center case series

Stephen C. Wong et al.

Summary: This retrospective case series highlights three cases of pediatric refractory autoimmune diseases that were treated with mesenchymal stem cell (MSC) therapy. The children showed improvement after the treatment, with two families believing their child was cured. While MSC therapy holds promise as an adjunctive treatment option for rheumatic diseases, further research and well-designed clinical trials are necessary for validation.

Chronic inflammation-induced senescence impairs immunomodulatory properties of synovial fluid mesenchymal stem cells in rheumatoid arthritis

Hyeon-Jeong Lee et al.

Summary: This study identified the presence of mesenchymal stem cell populations in the synovial fluid of rheumatoid arthritis patients, with long-term disease patients showing more cellular senescence, reduced immunomodulatory properties, and decreased anti-arthritic capacity in comparison to early disease patients. This suggests that chronic inflammatory milieu in rheumatoid arthritis affects the potential of mesenchymal stem cells.
