New 1,3,4-oxadiazoles linked with the 1,2,3-triazole moiety as antiproliferative agents targeting the EGFR tyrosine kinase

Summary: A series of 1,3,4-oxadiazole-1,2,3-triazole hybrids were designed and synthesized, and their antiproliferative activity against human cancer cell lines was evaluated. The most active compounds showed similar activity to erlotinib, and further studies revealed their ability to induce apoptosis and inhibit cancer cell growth.

Discovery of new pyrimido[5,4-c]quinolines as potential antiproliferative agents with multitarget actions: Rapid synthesis, docking, and ADME studies

Summary: A series of novel pyrimido[5,4-c]quinoline derivatives were synthesized and demonstrated to have in vitro antiproliferative activity. The compounds were evaluated for their inhibition of topoisomerase I, topoisomerase II alpha, CDK2, and EGFR. The results showed selective inhibitory activity against topoisomerase I and weak inhibitory activity against topoisomerase II alpha. Compound 30B exhibited the most potent anti-proliferative activity and moderate inhibition of CDK2. Compounds 30B, 28d, and 28c also showed significant inhibitory activity against EGFR.

Summary: The newly synthesized compound 12b exhibited excellent anticancer activity on MDA-MB-231 breast cancer cell line and was found to be safe for normal human bronchial epithelial cells. It induced apoptosis, depolarized mitochondrial membrane, arrested cells at sub-G2/M phase, and inhibited tubulin polymerization, supporting its effective anticancer potential.
