Reactive oxygen species (ROS)-responsive size-reducible nanoassemblies for deeper atherosclerotic plaque penetration and enhanced macrophage-targeted drug delivery

Jianhua He et al.

Summary: Nanoparticle-based therapeutics show potential for treating atherosclerosis, but the complex plaque microenvironment hinders their targeting ability. In this study, we developed reactive oxygen species (ROS)-responsive and size-reducible nanoassemblies to overcome this barrier. These nanoassemblies specifically accumulated in atherosclerotic plaque, rapidly disassembled in response to excess ROS, and released smaller nanoparticles for deeper penetration and targeted drug delivery. In vivo studies demonstrated that the nanoassemblies reduced plaque size, lipid deposition, macrophage content, and local inflammatory factor levels, while also alleviating systemic inflammation. Overall, the ROS-responsive and size-reducible nanoassemblies exhibit enhanced targeting ability and provide an effective approach for atherosclerosis therapy.

Self-oxygenation mesoporous MnO2 nanoparticles with ultra-high drug loading capacity for targeted arteriosclerosis therapy

Weidong Sun et al.

Summary: This study developed a MnO2-based nanomedicine to re-educate macrophages for targeted AS therapy. The nanomedicine exhibited catalase-mimic activity and high drug loading capacity, allowing it to effectively alleviate AS symptoms by repolarizing macrophages and promoting lipid efflux. In a mouse model, the nanosystem significantly enhanced drug accumulation in atherosclerotic lesions and achieved robust anti-AS efficacy.

In situ preparation of palladium nanoclusters in cerium metal-organic frameworks Ce-MOF-808, Ce-UiO-66 and Ce-BTC as nanoreactors for room temperature Suzuki cross-coupling reaction

Mostafa Farrag

Summary: Ultra-small thiolated palladium nanoclusters were immobilized inside cerium-organic frameworks, resulting in core-shell catalysts with high catalytic activity in Suzuki cross-coupling reactions. Among them, Pd-n@Ce-MOF-808 exhibited the highest turnover frequency and could be recycled multiple times without significant loss of activity. This study highlights the potential of palladium nanoclusters supported in MOFs for efficient catalysis in environmentally friendly conditions.

Atherosclerosis: Conventional intake of cardiovascular drugs versus delivery using nanotechnology - A new chance for causative therapy?

Jonas Groner et al.

Summary: Atherosclerosis, a chronic inflammatory disease, remains the leading cause of death in developed countries. Current cardiovascular drugs fail to address the root cause of the disease. Recent studies suggest that nanoparticulate carriers can enhance the bioavailability of statins and thiazolidinediones in atherosclerotic plaque, potentially improving their therapeutic effects.

Targeting inflammation in atherosclerosis - from experimental insights to the clinic

Oliver Soehnlein et al.

Summary: Inflammation plays a substantial role in atherosclerosis and targeting inflammatory pathways could provide a promising new avenue to prevent and treat the disease. Clinical studies have shown that modulation of inflammation can help prevent the clinical complications of atherosclerosis. Refining strategies for combatting inflammation in the disease is crucial for further progress in managing atherosclerotic risk.

The chemistry and applications of hafnium and cerium(iv) metal-organic frameworks

Zhigang Hu et al.

Summary: Metal-organic frameworks (MOFs) are formed by the coordination connection of organic linkers to metal clusters, with tunable ligands providing robust molecular porosity. Hafnium (Hf) and tetravalent cerium [Ce(iv)] MOFs have shown promise for practical applications, with a focus on their chemistry, topologies, synthetic strategies, and diverse applications. Future research will explore the development of Hf/Ce-MOFs, including fundamental design, defect engineering, and computational approaches for tailor-made applications.
