Oxygen regulates epithelial stem cell proliferation via RhoA-actomyosin-YAP/TAZ signal in mouse incisor

Keishi Otsu et al.

Summary: Oxygen plays a crucial role in regulating the proliferation of epithelial stem cells by maintaining slow-cycling cells in a relatively hypoxic microenvironment, and the upregulation of HIF1 alpha during hypoxia triggers a downstream signaling pathway that inhibits cell proliferation.

Spatiotemporal dynamics of SETD5-containing NCoR-HDAC3 complex determines enhancer activation for adipogenesis

Yoshihiro Matsumura et al.

Summary: The authors demonstrate that SETD5 forms a complex with NCoR-HDAC3 co-repressor to prevent histone acetylation of master adipogenic gene enhancers, while degradation of SETD5 leads to enhancer hyperacetylation and transition to an active state. Activation of the CDC20 co-activator of the ubiquitin ligase results in APC/C mediated degradation of SETD5, acting as a molecular switch that facilitates adipogenesis.

Ubiquitination-dependent and -independent repression of target genes by SETDB1 reveal a context-dependent role for its methyltransferase activity during adipogenesis

Ji Zhang et al.

Summary: Studies show that ubiquitination of SETDB1 complements its catalytic activity and silencing of endogenous retroviruses, but it is unclear if it is essential for silencing developmental genes. The ubiquitin-resistant K885A mutant of SETDB1 can still repress adipogenic genes due to compensation by other methyltransferases on chromatin modifications. This suggests that SETDB1 can repress its target genes through both ubiquitination-dependent and enzyme activity-independent mechanisms.
