Cross-grating phase microscopy (CGM):Insilicoexperiment (insilex)algorithm, noise and accuracy

Summary: Cross-grating phase microscopy (CGM) is a technique that combines a 2-dimensional diffraction grating and a regular camera sensor to achieve high-resolution imaging of the complex electric field of a light beam. In this article, a numerical algorithm is provided to simulate and observe the effects of various CGM experimental parameters. The algorithm is used to explain and quantify the influence of important CGM parameters on the noise, precision, and trueness of measurements. The aim of this work is to advance the applications of CGM in biomicroscopy and nanophotonics.

Summary: Quadriwave lateral shearing interferometry (QLSI) is a quantitative phase imaging technique that uses a diffraction grating to create an interferogram to retrieve the intensity and phase profiles of an incoming light beam. Despite its wide applications, efforts to disseminate this powerful and simple technique have been lacking. This article aims to popularize QLSI by describing its various aspects in the context of optical microscopy.

New insights into the diversity and evolution of the archaeal mobilome from three complete genomes of Saccharolobus shibatae

Summary: Saccharolobus shibatae, a hyperthermophilic and acidophilic archaeal species, serves as a host to spindle-shaped virus SSV1 and exhibits rich diversity of integrated mobile genetic elements. Analysis shows a complex network of MGE distributions among Sulfolobales species, involving horizontal spread and host switching over large phylogenetic distances. Additionally, a virus-to-plasmid transition has been characterized in this study.

Summary: The study overcame significant technical challenges in extremophile cell biology by using a newly developed imaging system that allowed observation and quantification of cell division dynamics in the thermoacidophilic crenarchaeon Sulfolobus acidocaldarius with unprecedented resolution. This system also revealed previously undescribed dynamic cell shape changes, cell motility, and cell-cell interactions, shedding new light on the high-temperature lifestyle of this organism.

Are bacteria claustrophobic? The problem of micrometric spatial confinement for the culturing of micro-organisms

Summary: The research demonstrates that culturing cells confined in microscale geometries might not be ideal using E. coli as a model, as spatial confinement can severely prevent the growth of many microorganisms. The inhibition of growth observed in some bacteria is determined to be due to fast dioxygen depletion caused by spatial confinement, rather than depletion of nutrients.
