Cyclic AMP signalling and glucose metabolism mediate pH taxis by African trypanosomes

Sebastian Shaw et al.

Summary: African trypanosomes exhibit social motility on semi-solid surfaces. The authors found that procyclic forms acidify their environment due to glucose metabolism, generating pH gradients that are sensed via cyclic AMP signaling. Parasite mutants defective in cAMP signaling are inhibited in fly infection.

Receptor-associated independent cAMP nanodomains mediate spatiotemporal specificity of GPCR signaling

Selma E. Anton et al.

Summary: Cells use independent cAMP nanodomains to send different signals, which form self-sufficient cell signaling units. Each nanodomain contains a highly localized cAMP pool that is protected from other receptors and cell compartments. The gradients of local cAMP concentrations define the size of each individual nanodomain. The coexistence of many such nanodomains allows a single cell to operate multiple independent cellular signals simultaneously.

Axelrod Symposium 2019: Phosphoproteomic Analysis of G-Protein-Coupled Pathways

Katharina Schleicher et al.

Summary: This review summarizes advanced imaging and proteomics techniques used to study compartmentalized cAMP signaling in eukaryotic cells, such as subcellularly targeted fluorescence resonance energy transfer sensors and proteomics analysis. The synergistic use of these techniques could potentially help in generating a detailed map of cAMP signalosomes and revealing new details of compartmentalized signaling.

Subcellular Organization of the cAMP Signaling Pathway

Manuela Zaccolo et al.

Summary: Recent developments in the field of cAMP signaling have highlighted the importance of cAMP compartmentalization and the complex network through which cAMP signals propagate. Understanding the spatial regulation of cAMP levels and selective activation of signaling branches within the network is crucial for determining specific functional outcomes and potential disease mechanisms. This knowledge offers new insights into cell physiology and opens avenues for innovative therapeutic interventions.

Liquid-liquid phase separation: Orchestrating cell signaling through time and space

Qi Su et al.

Summary: Cell signaling is a complex process that relies on the synergistic effects of multiple regulatory molecules for the accurate transmission of information. Liquid-liquid phase separation (LLPS) has been identified as a potentially important framework for organizing signaling molecules in cells. LLPS plays a crucial role in signal transduction by compartmentalizing signaling molecules, amplifying signals, and moderating signaling dynamics.

APEX2 Proximity Proteomics Resolves Flagellum Subdomains and Identifies Flagellum Tip-Specific Proteins in Trypanosoma brucei

Daniel E. Velez-Ramirez et al.

Summary: Trypanosoma brucei is the protozoan parasite responsible for sleeping sickness, and its flagellum plays critical roles in transmission and pathogenesis. APEX2-based proximity proteomics has been successfully used to examine the protein composition of T. brucei flagellum subdomains, allowing for the identification of unique proteomes for different regions of the flagellum.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.
