IDF Diabetes Atlas: Global, regional and country-level diabetes prevalence estimates for 2021 and projections for 2045

Hong Sun et al.

Summary: This study provides global, regional, and country-level estimates of diabetes prevalence and health expenditures for 2021 and projections for 2045. The global prevalence of diabetes is increasing and is expected to have the greatest relative increase in middle-income countries. Diabetes-related health expenditures are also projected to rise.

Lipotoxicity-induced circGlis3 impairs beta cell function and is transmitted by exosomes to promote islet endothelial cell dysfunction

Li Xiong et al.

Summary: The study found that under lipotoxic conditions, beta cell-derived exosomal circGlis3 was significantly upregulated, and levels of exosomal circGlis3 were also elevated in the serum of mouse models of diabetes and participants with type 2 diabetes. CircGlis3 participated in lipotoxicity-induced beta cell dysfunction in vitro and in vivo. Moreover, beta cell-derived exosomal circGlis3 could be transferred to islet ECs and reduce the cell viability, cell migration, and angiogenesis of islet ECs.

Pancreatic β cells control glucose homeostasis via the secretion of exosomal miR-29 family

Jing Li et al.

Summary: This study reveals that pancreatic beta cell-released exosomal miR-29 family members regulate hepatic insulin sensitivity and glucose homeostasis. High levels of free fatty acids stimulate the secretion of miR-29s, which in turn impact insulin signaling in the liver.

Long Non-Coding RNAs as Key Modulators of Pancreatic β-Cell Mass and Function

Livia Lopez-Noriega et al.

Summary: Despite the lack of a complete understanding of the pathogenesis of diabetes, the roles of long non-coding RNAs in beta-cell biology are crucial for insights into the disease mechanism. Long non-coding RNAs play essential roles in maintaining beta-cell mass and function.

Population-scale tissue transcriptomics maps long non-coding RNAs to complex disease

Olivia M. de Goede et al.

Summary: This study systematically characterized 14,100 lncRNA genes across 49 tissues for 101 complex genetic traits using GTEx project data, identifying 1,432 lncRNA gene-trait associations, 800 of which were not explained by neighboring protein-coding genes. The findings revealed associations between lncRNA and various diseases and traits.

Functional characterization of T2D-associated SNP effects on baseline and ER stress-responsive β cell transcriptional activation

Shubham Khetan et al.

Summary: Identifying causal variants at GWAS loci is crucial for understanding disease mechanisms, and this study used massively parallel reporter assays to pinpoint type 2 diabetes-associated variants and genetic mechanisms. The researchers revealed that multiple T2D-associated SNPs alter the activity of SINE-containing elements induced by ER stress, highlighting potential roles for repetitive elements in beta cell transcriptional stress response and T2D genetics.
