Lineage-selective super enhancers mediate core regulatory circuitry during adipogenic and osteogenic differentiation of human mesenchymal stem cells

Alterations in chromatin accessibility during osteoblast and adipocyte differentiation in human mesenchymal stem cells

Summary: In this study, we used ATAC-sequencing technology to observe variations in chromatin assembly during the early stages of MSC differentiation. Significant changes in the number and distribution of chromosome accessibility were found at different time points of adipogenic/osteogenic differentiation. Integration of ATAC-seq and RNA-seq data revealed a small proportion of overlapping genes. Analysis of these overlapped genes identified known biological functions related to adipogenic/osteogenic differentiation and unusual biological clustering during MSC differentiation.

Mapping the evolving landscape of super-enhancers during cell differentiation

Enhancers that emerge at early and late stages within temporally hierarchical super-enhancers are enriched with distinct transcription factors, indicating that the temporal order of establishment of elements within super-enhancers may be directed by underlying DNA sequence.
