The Lipid Droplet Knowledge Portal: A resource for systematic analyses of lipid droplet biology

Niklas Mejhert et al.

Summary: Lipid droplets (LDs) are organelles responsible for cellular lipid storage and play fundamental roles in energy metabolism and cell membrane homeostasis. Due to their relevance in lipid storage diseases, such as atherosclerosis, obesity, type 2 diabetes, and hepatic steatosis, there has been a growing need for a resource that combines systematically generated datasets on LD biology. The Lipid Droplet Knowledge Portal is an online platform that integrates high-confidence human, mouse, and fly data, providing a comprehensive resource for LD biology, including transcriptional profiles, proteomics, genome-wide screen phenotypes, and links to human genetics.

The CYTOLD and ERTOLD pathways for lipid droplet-protein targeting

Maria-Jesus Olarte et al.

Summary: In this review, the two pathways for direct LD protein localization are discussed: the CYTOLD pathway mediates protein targeting from the cytosol to LDs, and the ERTOLD pathway functions in protein targeting from the endoplasmic reticulum to LDs. The emerging principles for each targeting pathway in animal cells are described, along with highlighting open questions in the field.

The menage a trois of autophagy, lipid droplets and liver disease

Yasmina Filali-Mouncef et al.

Summary: Autophagic pathways intersect with lipid homeostasis to provide energy and building blocks essential for liver functions. Disruption of lipophagy regulation in fatty liver diseases, especially nonalcoholic steatohepatitis (NASH), may contribute to the development of hepatocellular carcinomas. This review focuses on the role of macroautophagy and macrolipophagy in NASH, highlighting the impact of inappropriate lipophagy on various types of liver cells.

Mechanisms of nonvesicular lipid transport

Karin M. Reinisch et al.

Summary: Lipids can be transported between cellular membranes via nonvesicular mechanisms, supporting biogenesis of rapidly expanding membranes or allowing targeted modulation of specific lipid levels. Nonvesicular transport likely occurs at membrane contact sites and involves lipid transport proteins, but many questions remain unanswered regarding the mechanisms involved.

Mechanism of lipid droplet formation by the yeast Sei1/Ldb16 Seipin complex

Yoel A. Klug et al.

Summary: The study revealed the mechanism of lipid droplet formation by the coordination of yeast Seipin Sei1 and its partner Ldb16. Sei1 assembles a homooligomeric ring in the ER to concentrate triacylglycerol, promoting Ldb16 stability and lipid droplet assembly.

Multiple C2 domain-containing transmembrane proteins promote lipid droplet biogenesis and growth at specialized endoplasmic reticulum subdomains

Amit S. Joshi et al.

Summary: The study reveals that transmembrane proteins MCTP1 and MCTP2 with multiple C2 domains are involved in lipid droplet (LD) formation and interact with seipin protein. These proteins can form microdomains to regulate LD biogenesis and size, as well as mediate ER-LD contacts.
