Discovery of a New CaMKII-Targeted Synthetic Lethal Therapy against Glioblastoma Stem-like Cells

Summary: The poor prognoses of patients with glioblastoma multiforme (GBM) are attributed to glioblastoma stem-like cells (GSCs) which induce drug resistance and rapid tumor recurrence. This study aims to explore a novel synthetic lethal therapy targeting calcium/calmodulin-dependent protein kinase II (CaMKII) for combatting GSCs. Through high-throughput drug combination screening, the combined treatment of CaMKII inhibitors and neurokinin 1 receptor (NK1R) inhibitors is found to exhibit chemical synthetic lethal effects on GSCs both in vitro and in vivo. It is proposed that this CaMKII-targeted combination therapy could be effective for treating GBM.

Physics of Brain Cancer: Multiscale Alterations of Glioblastoma Cells under Extracellular Matrix Stiffening

Summary: The biology and physics of glioblastoma are not completely understood, leading to limited effectiveness of current clinical therapy. Mechanical stress plays an important role in the development and malignancy of cancer, including glioblastoma. Alterations in the extracellular matrix (ECM) of the brain and the resulting stiffening of the ECM affect signaling activity and contribute to the aggressive behavior of glioblastoma cells. Mechanosensing signaling pathways, such as Hippo/YAP, CD44, and actin skeleton signaling, play a role in remodeling the cytoskeleton and influencing cellular properties. Hydrogels are used to mimic the stiffening of the brain ECM in vitro. Understanding these processes could potentially lead to new therapeutic approaches for glioblastoma.
