NAc-VTA circuit underlies emotional stress-induced anxiety-like behavior in the three-chamber vicarious social defeat stress mouse model

Guangjian Qi et al.

Summary: Emotional stress is a significant factor in psychiatric disorders, but the circuit mechanisms are unclear. This study used a 3C-VSDS mouse model and found that chronic emotional stress induced anxiety-like behavior and transient social interaction changes. Dopaminergic neurons in the VTA play a critical role in controlling this behavioral deficit, with their hyperactivity being involved in the anxiety-like behavior in an innate anxiogenic environment. Bidirectional modulation of the NAc-VTA circuit mimics or reverses the CES-induced anxiety-like behavior.

Summary: Livistolide A treatment improved memory deficits and cognitive decline in APP/PS1 Tg mice by stimulating the expression of alpha-secretase and reducing the generation of beta- and gamma-secretases. Inhibition of tau phosphorylation in the brains of Tg mice was also observed. The findings suggest that Livistolide A may be a candidate for AD treatment.

CDKL5 deficiency in forebrain glutamatergic neurons results in recurrent spontaneous seizures

Hong-Tao Wang et al.

Summary: The study demonstrates that CDKL5 cKO mice from Emx1- or CamK2 alpha-derived sources exhibit high-frequency spontaneous seizures resembling those in CDD patients, leading to sudden death in mice, while GABAergic neuron-specific CDKL5 deficiency does not generate such seizures. This emphasizes the potential of these mouse models for studying CDD-related seizures and therapeutic innovations.

Annexin-A1 SUMOylation regulates microglial polarization after cerebral ischemia by modulating IKKα stability via selective autophagy

Xing Li et al.

Summary: The study demonstrates that SUMOylated ANXA1 promotes the beneficial phenotype polarization of microglia by suppressing NF-κB activation and proinflammatory mediators. SUMOylated ANXA1 targets the IKK complex to enhance IKKα degradation and facilitates the interaction between IKKα and NBR1 for selective autophagy-mediated degradation, leading to improved neurological function in cerebral ischemia. Up-regulation of ANXA1 SUMOylation in microglia may provide therapeutic benefits for cerebral ischemia.

Pathology, Risk Factors, and Oxidative Damage Related to Type 2 Diabetes-Mediated Alzheimer's Disease and the Rescuing Effects of the Potent Antioxidant Anthocyanin

Muhammad Sohail Khan et al.

Summary: The pathology and neurodegeneration in type 2 diabetes-mediated Alzheimer's disease have common features such as brain atrophy, reduced cerebral glucose metabolism, and insulin resistance. Type 2 diabetes phenotypes like glucose dyshomeostasis and insulin resistance contribute to the progression of Alzheimer's disease pathology through mechanisms like amyloid-beta accumulation and overall neuroinflammation. Additionally, oxidative stress, mitochondrial dysfunction, and advanced glycation end products may link diabetes and Alzheimer's disease.

Dyrk1a gene dosage in glutamatergic neurons has key effects in cognitive deficits observed in mouse models of MRD7 and Down syndrome

Veronique Brault et al.

Summary: The perturbation of the excitation/inhibition balance can lead to neurodevelopmental diseases, including autism spectrum disorders, intellectual disability, and epilepsy. The DYRK1A gene plays a role in intellectual disability syndromes, with both loss-of-function and overexpression mutations having different effects. In postnatal glutamatergic neurons, Dyrk1a dosage does not impact locomotor activity or working memory but is involved in long-term explicit memory.

The mTOR/NF-κB Pathway Mediates Neuroinflammation and Synaptic Plasticity in Diabetic Encephalopathy

Ting Xu et al.

Summary: Diabetic encephalopathy, characterized by neuroinflammation and aberrant synaptogenesis, involves the mTOR and NF-kappa B signaling pathways. Inhibiting mTOR and NF-kappa B can reduce inflammatory cytokines expression and increase synaptic proteins, leading to the improvement of cognitive decline in diabetic models. This study suggests that targeting the mTOR/NF-kappa B pathway could be a potential therapeutic strategy for treating diabetic encephalopathy and related cognitive impairments.
