Pyrimidine inhibitors synergize with nucleoside analogues to block SARS-CoV-2

Summary: This study screened and validated 122 drugs with antiviral activity and selectivity against SARS-CoV-2 out of approximately 18,000 drugs. Among them are the approved antivirals remdesivir and molnupiravir. The study also found that combining pyrimidine biosynthesis inhibitors with antiviral nucleoside analogues synergistically inhibits SARS-CoV-2 infection.

Summary: Global analysis showed low susceptibility of influenza viruses to neuraminidase inhibitors (NAIs) and the polymerase acidic (PA) inhibitor baloxavir. NA-H275Y substitution in A(H1N1)pdm09 viruses conferred high resistance to oseltamivir and peramivir. Baloxavir showed reduced susceptibility in a small percentage of viruses, with Japan having a higher rate. Overall, NAIs and baloxavir remain appropriate choices for treating influenza virus infections.

Inhibition of DHODH increases the incorporation of NHC into viral RNA, leading to defective virus genomes. The combination of NHC and DHODH inhibitors may improve therapy options for COVID-19.

The Synergism between DHODH Inhibitors and Dipyridamole Leads to Metabolic Lethality in Acute Myeloid Leukemia

Summary: In this study, we investigated the pro-apoptotic and pro-differentiating activity of MEDS433, an inhibitor of Dihydroorotate Dehydrogenase, in acute myeloid leukemia (AML). We found that the combination of MEDS433 and dipyridamole induced metabolic lethality and myeloid differentiation in all AML models, with limited toxicity on non-AML cells. This synergistic effect was confirmed on AML primary cells with different genetic backgrounds, suggesting potential clinical relevance.

The antiviral activity of MEDS433 was reversed by the addition of exogenous uridine or orotate, confirming hDHODH as the specific target in hCoVs-infected cells.
