Summary: The emergence of multidrug resistance has become a global environmental safety concern. This study investigated the effect of heavy metals on antibiotic resistance by analyzing the alteration of resistance phenotype under Cu2+ and Zn2+ exposure. The results showed that heavy metals can induce bacteria to develop antibiotic resistances that were not previously available, and they may enhance antibiotic resistance by altering the expression of efflux pump genes.

Molecular mechanism of antibiotic resistance induced by mono- and twin-chained quaternary ammonium compounds

Yin Jia et al.

Summary: The usage of quaternary ammonium compounds (QACs) as disinfectants has increased dramatically during the COVID-19 pandemic, potentially leading to accelerated antibiotic resistance. Long-term exposure to subinhibitory level QACs can lead to multidrug resistance, but the role of mutagenesis in resistance evolution is not clear. This study found that E. coli adapted differently to benzalkonium chloride (BAC) and didecyldimethylammonium chloride (DDAC), with DDAC exposure resulting in higher antibiotic resistance and more mutations. The mutations induced by QACs potentially contributed to high multi-drug resistance. The reversible and irreversible phenotypic resistance induced by subinhibitory and inhibitory concentrations of QACs, respectively, highlight the importance of understanding the mechanisms and risks of different QACs in the environment.

Beyond horizontal gene transfer: the role of plasmids in bacterial evolution

Jeronimo Rodriguez-Beltran et al.

Summary: Plasmids play a vital role in bacterial ecology and evolution by mobilizing accessory genes through horizontal gene transfer. Recent studies show that plasmids, kept at multiple copies per cell, create islands of polyploidy in bacterial genomes, leading to evolution governed by different rules than those affecting chromosomal genes. Plasmids may accelerate bacterial evolution by promoting the evolution of plasmid-encoded genes and enhancing the adaptation of their host chromosomes.

Potential of 1-(1-napthylmethyl)-piperazine, an efflux pump inhibitor against cadmium-induced multidrug resistance in Salmonella enterica serovar Typhi as an adjunct to antibiotics

Ujjwal Jit Kaur et al.

Summary: This study focused on elucidating the inhibition of antibiotic efflux mechanism in cadmium-adapted Salmonella Typhi Ty2 cells. The research found that efflux pump inhibitors (EPIs) could effectively inhibit even the overexpressed efflux pumps, resulting in a decrease in the effective concentration of antibiotics. This suggests that the combination of EPIs with antibiotics could be a potential strategy for combating metal-induced antibiotic resistance.
