Snake venom gene expression is coordinated by novel regulatory architecture and the integration of multiple co-opted vertebrate pathways

Summary: Understanding the evolution of regulatory mechanisms is crucial for comprehending the origin of novel phenotypes. Snake venom systems serve as a valuable and easily manageable model for testing hypotheses about the evolution of regulatory networks, although the regulatory mechanisms underlying venom production remain unclear. In this study, functional genomics approaches were employed to investigate the venom regulatory architecture in the prairie rattlesnake. The study identified cis-regulatory sequences, trans-regulatory transcription factors, and integrated signaling cascades involved in the regulation of snake venom genes. The findings suggest that conserved vertebrate pathways were co-opted to regulate snake venom, and the co-option was likely facilitated by transposable elements. The conservation patterns of snake venom gene enhancers and the features of chromatin structure involved in venom regulation further highlight the early origins and lineage-specific adaptations that have occurred during the evolution of venom regulatory architecture.
