State of the art of nuclear transfer technologies for assisting mammalian reproduction

Andres Gambini et al.

Summary: Nuclear transfer techniques are important tools for studying epigenetic reprogramming, nucleus-cytoplasm compatibility, pluripotency state, and genetic preservation or edition in animals. When the nuclear donor cell is an oocyte, egg, or a zygote, these techniques have clinical relevance for patients with repeated failures in ART.

Extracellular vesicles from follicular fluid may improve the nuclear maturation rate of in vitro matured mare oocytes

Julia Gabrys et al.

Summary: This study aims to investigate the effect of follicular fluid extracellular vesicles (ffEVs) on the in vitro maturation (IVM) of equine oocytes. The supplementation of ffEVs from small ovarian follicles was found to significantly improve the IVM rate of mare oocytes, indicating the important role of ffEVs in this process and their potential to enhance equine assisted reproductive techniques (ART) development.

Success rate in a clinical equine in vitro embryo production program

A. Claes et al.

Summary: In vitro embryo production (IVEP) using Ovum Pick-Up (OPU) and Intracytoplasmic Sperm Injection (ICSI) has become a popular breeding technique for Warmblood mares. It offers a solution for various reproductive issues and has a high success rate. The average number of antral follicles aspirated per session was 25.9, resulting in an average of 13.8 immature oocytes. The likelihood of pregnancy after transfer of a cryopreserved thawed IVP blastocysts in 2021 was 77.7%.

Successful in vitro fertilization in the horse: production of blastocysts and birth of foals after prolonged sperm incubation for capacitation†

Matheus R. Felix et al.

Summary: This study demonstrates that pre-incubation of fresh equine sperm with penicillamine, hypotaurine, and epinephrine can greatly improve the success rates of in vitro fertilization and the production of viable blastocysts. The use of extended semen and 22 hours of pre-incubation resulted in high rates of fertilization and blastocyst formation. This is the first report of successful standard in vitro fertilization in horses and the production of blastocysts and foals through this technique.

Correlation between Pre-Ovulatory Follicle Diameter and Follicular Fluid Metabolome Profiles in Lactating Beef Cows

Casey C. Read et al.

Summary: In small beef cows, induced ovulation of small pre-ovulatory follicles leads to reduced pregnancy rates, embryo survival, and quality, potentially due to insufficient glucose metabolism capacity of cumulus cells in smaller follicles. The study explored the correlation between pre-ovulatory follicle diameter and follicular fluid metabolome profiles, identifying 18 metabolites significantly positively correlated with follicle diameter. This suggests altered glucose and amino acid metabolism may contribute to decreased developmental competence of oocytes when small pre-ovulatory follicles undergo induced ovulation.
