The data suggest extensive promiscuity of initial ligand binding and large-scale floating motions of GPCR transmembrane helices in the lipid bilayer.

This site-specific F-19 NMR labeling method is a powerful tool for decoding the mechanism of GPCR allosteric modulation.

Delineating the conformational landscape of the adenosine A2A receptor during G protein coupling

Summary: This study used fluorine nuclear magnetic resonance spectroscopy to identify key functional states of the adenosine A(2A) receptor complexed with heterotrimeric G protein in a phospholipid membrane environment. The findings revealed an ensemble consisting of inactive states, a G-protein-bound activation intermediate, and nucleotide-free states associated with agonist-driven activation. The crucial role of the G beta gamma subunit in facilitating ligand-dependent allosteric transmission was demonstrated.

Crystal Structure and Subsequent Ligand Design of a Nonriboside Partial Agonist Bound to the Adenosine A2A Receptor

Summary: In this study, the crystal structure of an engineered human adenosine A(2A) receptor bound to a partial agonist was determined and compared to structures bound to a full agonist or antagonist/inverse agonist. Interaction between the partial agonist and amino acids in the ligand binding pocket led to the development of a library of derivatives with varied activities. Some derivatives showed partial agonist properties while others acted as inverse agonists, with additional computational docking studies providing insights into this behavior.

Expression and Purification of Yeast-derived GPCR, Gα and Gβγ Subunits for Structural and Dynamic Studies

Summary: In recent years, significant progress has been made in the structural biology of GPCRs and G proteins, with over 70 GPCRs structures resolved, but challenges remain in terms of the quantity of receptors and proteins required for conformational transition and dynamics studies. The Pichia pastoris expression system has shown promise in providing an effective and affordable solution for GPCR preparation, with advantages that other eukaryotic expression systems cannot match. Developed protocols for expressing and purifying GPCRs in this system are expected to advance studies on conformational transitions and dynamics of GPCRs and their complexes.
