Summary: This study used cryo-EM to determine the three-dimensional structure of human lysosomal α-L-fucosidase, revealing its structural characteristics, the architecture of the catalytic center, and the location of natural population variations and disease-causing mutations. Furthermore, the study identified the catalytic acid/base and its shift from the canonical glutamate residue. These findings provide important insights into the function of FucA1 in both health and disease.
