Orderly Assembled, Self-Powered FRET Flares for MicroRNA Imaging in Live Cells

Summary: This study aimed to develop an integrated amplification platform for detecting intracellular miRNA, termed self-powered FRET flares (SPF), which achieved target recycling and signal amplification under the trigger of intracellular miRNA-21 without the need for auxiliary additives. By utilizing AS1411 aptamer to target specific cancer cells and facilitating cell internalization of assembly DNA nanostructures, SPF demonstrated rapid response to miRNA-21 and improved detection sensitivity compared to previous FRET flares without amplification, showing promise for diverse tasks in biological and medical applications.

DNAzyme cascade circuits in highly integrated DNA nanomachines for sensitive microRNAs imaging in living cells

Flap Endonuclease 1-Assisted DNA Walkers for Sensitively and Specifically Sensing ctDNAs

Summary: A DNA walker assisted by flap endonuclease 1 (FEN 1) has been proposed to achieve mutant biosensing with high specificity and sensitivity. The system is capable of discriminating single-base mutant DNA targets from wild-type DNA, and can detect DNA targets as low as 1 fM and mutation abundance as low as 0.1%. Detection of epidermal growth factor receptor (EGFR) L858R mutation on cell-free DNA samples showed results consistent with next-generation sequencing, indicating the potential of the DNA walker for liquid biopsy.

A self-assembled DNA nanostructure as a FRET nanoflare for intracellular ATP imaging
