Discovery of In Vivo Active Sphingosine-1-phosphate Transporter (Spns2) Inhibitors

Russell Fritzemeier et al.

Summary: In this study, the researchers identified a novel Spns2 inhibitor called 16d, which effectively inhibits the release of S1P. In addition, administration of 16d in mice and rats replicated the phenotype observed in Spns2-deficient mice. This study provides a new research tool for further understanding the biological function of Spns2 and evaluating its potential as a drug target.

Endothelial S1P1 Signaling Counteracts Infarct Expansion in Ischemic Stroke

Anja Nitzsche et al.

Summary: This study explores the role of endothelial cell S1P(1) in the naive and ischemic brain as a potential target for cerebrovascular therapy. Endothelial S1P(1) plays a critical vascular protective role in the mouse brain, sustaining endothelial functions and expanding during ischemia. Disruption of this pathway exacerbates brain injury in ischemic stroke models, highlighting the importance of S1P signaling in maintaining perfusion and microvascular patency in the ischemic penumbra.

Functional characterization of the dural sinuses as a neuroimmune interface

Justin Rustenhoven et al.

Summary: The CNS-derived antigens in the cerebrospinal fluid accumulate around the dural sinuses and are captured by local antigen-presenting cells to be presented to T cells, promoting tissue resident phenotypes and effector functions within the dural meninges. The dural sinuses play a critical role as a neuroimmune interface, facilitating the surveillance of brain antigens under steady-state conditions.

Vagus nerve stimulation activates two distinct neuroimmune circuits converging in the spleen to protect mice from kidney injury

Shinji Tanaka et al.

Summary: The study found that VNS can protect the kidneys from ischemia-reperfusion injury by stimulating vagus nerve fibers and identified the related neural circuits; this is crucial for the clinical application of VNS for protection from acute kidney injury.
