Chronic exposure to realistic concentrations of metformin prompts a neurotoxic response in Danio rerio adults

Chronic exposure to environmentally relevant concentrations of guanylurea induces neurotoxicity of Danio rerio adults

Summary: Recent studies have shown that guanylurea (GUA) has adverse effects on the growth and development of fish, induces oxidative stress, and disrupts the levels and expression of several genes, metabolites, and proteins related to fish fitness. The aim of this study was to investigate the potential neurotoxic effects of environmentally relevant concentrations of GUA in fish. The results demonstrated that chronic exposure to GUA altered the swimming behavior of the fish, increased oxidative damage, and inhibited acetylcholinesterase activity. Furthermore, GUA altered the expression of several genes related to antioxidant defense, apoptosis, AMPK pathway, and neuronal communication in the brain of the fish.

Metformin-Induced Epigenetic Toxicity in Zebrafish: Experimental and Molecular Dynamics Simulation Studies

Summary: The study investigated the effects of metformin exposure on tissue accumulation, gene expression, and global DNA methylation in zebrafish. Metformin accumulated in the gills, gut, and liver of zebrafish, with the liver being the main site of global DNA methylation. Metformin also induced abnormal gene expression in the liver due to hydrogen bond formation. The molecular dynamics simulation allowed for the mechanistic determination of metformin-induced conformational changes to DNA methyltransferase 1.
