AOP Key Event Relationship report: Linking decreased retinoic acid levels with disrupted meiosis in developing oocytes

Monica Kam Draskau et al.

Summary: The Adverse Outcome Pathway (AOP) is an emerging tool in regulatory toxicology that shows simplified cause-effect relationships between stressors and toxicity outcomes. Key Event Relationships (KERs) are the core building blocks within the AOP-Knowledge Base, representing causal relationships between two Key Events. This study presents a case supporting the recognition of KERs as the core building blocks and describes a KER linking decreased atRA levels with disrupted meiotic entry of oogonia.

A Pragmatic Approach to Adverse Outcome Pathway Development and Evaluation

Terje Svingen et al.

Summary: The AOP framework organizes scientific knowledge to infer cause-effect relationships between stressor events and toxicity outcomes, supporting chemical safety assessment and regulatory toxicology. However, developing robust AOPs requires substantial work and time, leading to a proposed more pragmatic approach for AOP development.
