Predictive Performance of Next Generation Physiologically Based Kinetic (PBK) Model Predictions in Rats Based on In Vitro and In Silico Input Data

Ans Punt et al.

Summary: The present study assesses the predictive performance of a minimal generic rat physiologically based kinetic (PBK) model to predict peak plasma concentrations (C-max) upon single oral dosing based on in vitro and in silico input data. The best performance is obtained by parameterizing the hepatic clearance, calculating partition coefficients, and using in silico calculated fraction unbound plasma and P-app values. The study provides relevant insights into the predictive performance of the model.

Integrating in vitro chemical transplacental passage into a generic PBK model: A QIVIVE approach

Styliani Fragki et al.

Summary: With the increasing application of cell culture models, the quantitative extrapolation of effective dose from in vitro to in vivo (QIVIVE) becomes important in predicting chemical safety. This study investigated the transfer of substances across the placental barrier using the BeWo cell assay and incorporated the results into a Physiologically Based Kinetic (PBK) model. The approach allowed for a comparison of in vitro dose-response characteristics with in vivo rat developmental toxicity data.

In Silico Prediction of Input Parameters for Simplified Physiologically Based Pharmacokinetic Models for Estimating Plasma, Liver, and Kidney Exposures in Rats after Oral Doses of 246 Disparate Chemicals

Yusuke Kamiya et al.

Summary: Recent computational models have been developed to estimate concentrations of industrial chemicals in rats, utilizing machine learning algorithms and a variety of chemical properties. By incorporating these estimated parameters into PBPK models, internal exposures in rats can be predicted and potential risks can be assessed. The correlation between traditionally determined and in silico estimated input parameters in rat PBPK models demonstrates the potential of using forward dosimetry to estimate plasma/tissue concentrations of drugs and chemicals.

Paving the way for application of next generation risk assessment to safety decision-making for cosmetic ingredients

Summary: NGRA is an exposure-led, hypothesis-driven approach that has the potential to support animal-free safety decision-making. However, it is still in its early stages and further case studies are needed to determine if safety decisions are sufficiently protective and not overly conservative. Efforts should also be focused on developing frameworks to guide performance and reporting.

High-throughput PBTK models for in vitro to in vivo extrapolation

Miyuki Breen et al.

Summary: This review discusses the use of generic physiologically based toxicokinetic (PBTK) models and high-throughput PBTK modeling for in vitro-in vivo extrapolation (IVIVE). The 'httk' tool is highlighted as a useful resource for chemical risk assessors to support rapid screening and prioritization, as well as to provide TK modeling for large numbers of chemicals using limited data. The modular and open-source design of httk allows for continuous improvement by a broad user community to facilitate rapid hazard assessments.
