Cross-linking mass spectrometry reveals the structural topology of peripheral NuRD subunits relative to the core complex

Cornelia G. Spruijt et al.

Summary: The study investigates the relative orientation of different subunits within the NuRD complex using multiple cross-link IP mass spectrometry experiments, confirming that the core of the complex consists of MTA, RBBP, and HDAC proteins. It also reveals interactions between CDK2AP1 and CDK2AP2 with the NuRD complex, shedding light on the topology of peripheral NuRD subunits relative to the core complex. The data provide a more detailed understanding of the assembly and structure of the NuRD complex.

UTRN inhibits melanoma growth by suppressing p38 and JNK/c-Jun signaling pathways

Sitong Zhou et al.

Summary: Utrophin, a tumor suppressor gene, showed decreased expression in melanoma and was associated with poorer prognosis. Experimental evidence supported that up-regulated utrophin expression could inhibit melanoma cell proliferation. The inhibition of melanoma growth by utrophin was mediated through p38 and JNK1/c-Jun pathways.

The DBSAV Database: Predicting Deleteriousness of Single Amino Acid Variations in the Human Proteome

Jimin Pei et al.

Summary: The study aimed to enhance the performance of DeepSAV by using expanded datasets, more features, and neural network optimization. It was found that multiple sequence alignments built from vertebrate-level orthologs yield better prediction results compared to those built from mammalian-level orthologs, and optimal performance was achieved with a sequence identity cutoff of 50%. The new version of DeepSAV exhibits the best performance among standalone predictors of deleterious effects of SAVs.

Structure of the FA core ubiquitin ligase closing the ID clamp on DNA

Shengliu Wang et al.

Summary: The Fanconi anemia pathway relies on the FA core complex for repairing DNA interstrand crosslinks, which is achieved through an asymmetric assembly preventing inhibition of the recruitment of ubiquitin conjugating enzyme. The complex sequentially ubiquitinates FANCD2 and FANCI at a single active site, preventing clamp opening after release. The cryo-EM structures shed light on the remodeling process of the FANCI-FANCD2 DNA clamp, essential for DNA interstrand crosslink repair.

Mechanisms of BRCA1-BARD1 nucleosome recognition and ubiquitylation

Qi Hu et al.

Summary: The BRCA1-BARD1 tumor suppressor complex plays a crucial role in DNA repair through homologous recombination. The complex is recruited to damaged chromatin and catalyzes ubiquitylation of histones, with BARD1's ankyrin repeat and BRCT domains binding to nucleosomal histones, DNA, and monoubiquitin attached to H2A. This mechanism involves a regulatory crosstalk between BRCA1-BARD1 and H2A, promoting ubiquitylation and opposing 53BP1 to promote homologous recombination.
