The KRAB Domain of ZNF10 Guides the Identification of Specific Amino Acids That Transform the Ancestral KRAB-A-Related Domain Present in Human PRDM9 into a Canonical Modern KRAB-A Domain

Peter Lorenz et al.

Summary: This study analyzed aKRAB sequences from different subgroups in the animal kingdom and defined ancestral subdomains for KRAB-A and KRAB-B. It identified the necessary amino acid changes that transform the inactive aKRAB into a functional mKRAB domain. The study revealed the protein-protein interaction code between different KRAB domains and TRIM28, providing basic insights into structure-function relationships.

AF2Complex predicts direct physical interactions in multimeric proteins with deep learning

Mu Gao et al.

Summary: This study introduces a method called AF2Complex, which uses neural network models developed for individual protein sequences in AlphaFold2 to accurately predict the structure of multimeric protein complexes without retraining. The method achieves higher accuracy in predicting protein-protein interactions compared to common approaches and shows significant improvement over the AlphaFold version for multimeric proteins. The effectiveness of AF2Complex is validated on benchmark sets and the E. coli proteome, and high-confidence models of sought-after assemblies in the cytochrome c biogenesis system I are presented.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.
