Senescence and impaired DNA damage responses in alpha-synucleinopathy models

Ye-Seul Yoon et al.

Summary: Excessive expression of Parkinson's protein impairs DNA repair and leads to cellular senescence. The accumulation of DNA double-strand breaks and cellular senescence are intermediaries of alpha-synuclein-induced pathogenesis in Parkinson's disease.

Phosphorylation of the aggregate-forming protein alpha-synuclein on serine-129 inhibits its DNA-bending properties

Sydney E. Dent et al.

Summary: In this study, we found that both alpha-synuclein (aSyn) and its phosphorylated form (pSyn) can bind DNA in a DNA length-dependent manner, but without specificity for DNA sequence. We also observed that the phosphorylation of serine-129 reduces the ability of aSyn to bind and bend DNA. These findings suggest that the ability of aSyn to interact with DNA and modulate DNA metabolism may be regulated by phosphorylation.

APE1/Ref-1 Role in Inflammation and Immune Response

Thais Teixeira Oliveira et al.

Summary: APE1 is a multifunctional enzyme that is essential for maintaining cellular homeostasis and regulating immune response. It plays important roles in cell signaling, senescence, and inflammatory pathways, and is involved in the pathogenesis of various diseases, including cancer and neurological disorders. APE1 inhibitors have potential therapeutic uses, particularly in infectious and immune diseases.

Neuropathological consensus criteria for the evaluation of Lewy pathology in post-mortem brains: a multi-centre study

Summary: Several staging systems exist for the neuropathological diagnosis of LBD, with varying levels of inter-rater reliability. The LPC system, based on a dichotomous approach for scoring LP, showed good reproducibility and allowed for the classification of all cases into distinct categories. It may serve as a reliable and useful standard future approach for the post-mortem evaluation of LP.

Astrogliosis and sexually dimorphic neurodegeneration and microgliosis in the olfactory bulb in Parkinson's disease

Alicia Flores-Cuadrado et al.

Summary: Hyposmia is an early symptom of Parkinson's disease, with male gender being a risk factor. The reduction of olfactory bulb volume is controversial, but it is considered an early and preferential site for alpha-synucleinopathy. Neurodegeneration and microgliosis differences are only present in men, suggesting sex differences in neurodegeneration in the human olfactory bulb in Parkinson's disease.

Coping with RNA damage with a focus on APE1, a BER enzyme at the crossroad between DNA damage repair and RNA processing/decay

Matilde Clarissa Malfatti et al.

Summary: Interest in RNA damage as a novel threat associated with human pathologies is growing, yet knowledge on damaged RNA recognition, repair, processing, and decay remains limited. APE1, initially identified as a key enzyme in DNA base excision repair, has also been found to play crucial roles in gene expression control, RNA metabolism, and regulation of oncogenic miRNAs. Recent findings suggest APE1's involvement in RNA processing activities, highlighting the need for detailed understanding of damaged RNA decay in human cells to comprehend multiple pathologies.

α-synuclein aggregates induce c-Abl activation and dopaminergic neuronal loss by a feed-forward redox stress mechanism

Soumitra Ghosh et al.

Summary: Oxidative stress and alpha-synuclein aggregation play significant roles in driving neurodegeneration in Parkinson's disease. The protein kinase c-Abl is identified as a potential link between these pathogenic factors. The use of N-acetyl cysteine shows promise in suppressing oxidative stress and neuronal damage.

DNA Repair Protein APE1 Degrades Dysfunctional Abasic mRNA in Mitochondria Affecting Oxidative Phosphorylation

Arianna Barchiesi et al.

Summary: APE1 is a versatile protein involved in DNA repair, gene expression regulation, and RNA metabolism in mitochondria. Loss of APE1 leads to accumulation of damaged mitochondrial mRNA, impairing protein translation and mitochondrial respiration efficiency. The data demonstrate that APE1 plays a crucial role in maintaining mitochondrial well-being.

Heat Shock Protein 70 as a Sex-Skewed Regulator of α-Synucleinopathy

Tarun N. Bhatia et al.

Summary: The study revealed gender differences in the role of Hsp70 in proteinopathic disorders, with exogenous Hsp70 reducing alpha-synuclein inclusions in cells of both sexes. Intranasal delivery of eHsp70 in male mice decreased disease progression, while no eHsp70 uptake into the brain was observed in females. Inducible Hsp70 expression was higher in amygdala tissues of male subjects, while women expressed higher levels of amygdalar Hsp70 regardless of disease status.
