Targeting cell-matrix interface mechanobiology by integrating AFM with fluorescence microscopy

Molecular Engineering of Pericellular Microniche via Biomimetic Proteoglycans Modulates Cell Mechanobiology

Summary: Molecular engineering using synthetic mimics of native matrix molecules can modulate the mechanical properties of cellular microenvironment and influence cell mechanobiology. This study demonstrated the use of biomimetic proteoglycans (BPGs) to engineer the micromechanics of the pericellular matrix (PCM) in cartilage, leading to enhanced cellular mechanotransduction. The interactions between BPGs and the native PCM were facilitated by the biomimetic ultrastructure of BPGs, and showed potential for improving tissue regeneration and disease modification in various cell types.

AFM and Fluorescence Microscopy of Single Cells with Simultaneous Mechanical Stimulation via Electrically Stretchable Substrates

Summary: A novel experimental set-up has been developed to simultaneously apply static and dynamic deformations to adherent cells in culture, visualize cells under fluorescence microscopy, and measure the mechanical properties of the cells using atomic force microscopy nanoindentation. The device relies on a dielectric elastomer film that can be electro-actuated and controlled to obtain specific deformations. Proof of performance on fibroblasts shows a reproducible increase in cell elastic modulus in response to applied uniaxial stretch. This platform allows real-time investigation of single cell mechano-transduction phenomena with sub-cellular spatial resolution.
