Summary: People of recent sub-Saharan African ancestry are more prone to kidney failure, largely due to two coding sequence variants in the APOL1 gene. These variants, G1 and G2, are known to be toxic gain-of-function variants, causing diseases like FSGS when inherited in two copies. Studies with transgenic mice have shown that disease risk largely depends on the level of risk variant APOL1 expression.
