Apolipoprotein L1 High-Risk Genotypes and Albuminuria in Sub-Saharan African Populations

Jean-Tristan Brandenburg et al.

Summary: This study investigated the prevalence of APOL1 risk alleles and their association with kidney disease in different regions of Africa. The results showed that APOL1 risk variants were significantly associated with albuminuria but not eGFR decline. The frequencies of high-risk genotypes differed between different regions of Africa.

Kidney-disease-associated variants of Apolipoprotein L1 show gain of function in cation channel activity

Summary: ApoL1 variants are linked to increased risk of progressive kidney diseases among people of African ancestry. These variants show significantly increased cation-selective ion permeability, suggesting that enhanced cation permeability may contribute to the development of these kidney diseases.

Summary: This study identified six recent African ancestry patients who developed COVID-19-associated AKI with podocytopathy, collapsing glomerulopathy, or both. Genetic testing in three patients confirmed high-risk APOL1 genotypes, suggesting a potential link between glomerular disease in COVID-19 patients and high-risk APOL1 genotypes.

APOL1 risk variants affect podocyte lipid homeostasis and energy production in focal segmental glomerulosclerosis

Mengyuan Ge et al.

Summary: The study reveals that the expression of the APOL1 risk variant G1 exacerbates proteinuria and kidney function damage in an FSGS mouse model, leading to lipid accumulation in kidney cortices and impaired renal function. Additionally, expression of APOL1 G1/G2 in human urinary podocytes is associated with increased cellular triglyceride content and mitochondrial dysfunction.

Summary: African-Americans have a three-fold higher rate of chronic kidney disease compared to European-Americans, largely attributed to genetic variants in the APOL1 gene. Despite many mechanisms reported in cell culture models, few have been proven to be effective in transgenic models.

Coiled-coil binding of the leucine zipper domains of APOL1 is necessary for the open cation channel conformation

Charles Schaub et al.

Summary: APOL1 acts as a channel-forming effector of innate immunity, with different variants providing protection against certain parasite infections. However, G1 and G2 variants may also increase the risk of chronic kidney disease. The structural determinants of cation channel formation by APOL1 include the C-terminal leucine zipper domain, which is essential for channel formation and lysis of parasites and mammalian cells.

Novel Human Podocyte Cell Model Carrying G2/G2 APOL1 High-Risk Genotype

Pepe M. Ekulu et al.

Summary: Apolipoprotein L1 (APOL1) high-risk genotypes G1 and G2 increase the risk of non-diabetic kidney diseases in the African population. This study found that induction of APOL1 expression led to podocyte dysfunction in both G2/G2 and G0/G0 podocytes, with G2/G2 cells showing more altered features. The human APOL1 G2/G2 podocyte cell model provides a useful tool for understanding APOL1-induced podocyte injury and cellular functions.
