TNF-mediated alveolar macrophage necroptosis drives disease pathogenesis during respiratory syncytial virus infection

Leonardo Duarte Santos et al.

Summary: Respiratory syncytial virus (RSV) infection triggers necroptosis in macrophages, affecting RSV clearance and contributing to lung injury. Depletion of alveolar macrophages protects mice from weight loss and reduces viral loads in the lungs during RSV infection. Targeting TNF and/or the necroptotic machinery may be valuable therapeutic approaches to reduce respiratory morbidity caused by RSV infection in young children.

Optimization of an air-liquid interface in vitro cell co-culture model to estimate the hazard of aerosol exposures

Rui-Wen He et al.

Summary: Inhalation exposure to aerosol contaminants can lead to respiratory health problems. To mimic long-term exposure for toxicity testing, lung epithelial cells must be maintained under ALI conditions and co-cultured with macrophages. Calu-3 cells maintain monolayer structure and tight junctions, making them ideal for co-culturing with macrophages for increased sensitivity in responses to aerosol exposure.

Alveolar-like Macrophages Attenuate Respiratory Syncytial Virus Infection

Barbara N. Porto et al.

Summary: The study demonstrates that ALMs generated from pluripotent stem cells can prevent RSV infection by phagocytosis of virus particles and releasing an antiviral factor. ALMs protect mice from virus-induced weight loss and reduce lung inflammation in experiments, indicating their potential prophylactic role in RSV infection.

Inflammatory epithelial cytokines after in vitro respiratory syncytial viral infection are associated with reduced lung function

Wenming Duan et al.

Summary: The study found that cystic fibrosis epithelial cells have an abnormal response to RSV, leading to increased viral replication and exaggerated inflammatory response, which may ultimately exacerbate airway inflammation and decrease lung function.
