PI3K-driven HER2 expression is a potential therapeutic target in colorectal cancer stem cells

Laura Rosa Mangiapane et al.

Summary: In colorectal cancer stem cells, high HER2 expression levels are associated with activation of the PI3K/AKT pathway. The targeting of HER2 in combination with PI3K and MEK inhibitors induces CR-CSC death and regression of tumor xenografts, including those with specific mutations. Triple targeting of PI3K, HER2, and MEK is necessary to overcome resistance to anti-EGFR therapies.

Extraction and detection of morin from Sanghuangporus lonicericola by magnetic molecularly imprinted polymers coupled with HPLC analysis

Wei Zhang et al.

Summary: In this study, morin magnetic molecularly imprinted polymers (Morin-MMIPs) were synthesized using magnetic nanoparticles and surface molecularly imprinted technology. The polymers exhibited superparamagnetism and extraction selectivity, allowing for the separation of morin from complex matrices in the presence of an external magnetic field. Characterization of the polymers showed their microstructure and functional group composition, as well as their magnetic properties. The adsorption capacity of MMIPs was significantly higher than that of MNIPs. The method reported in this study provided a reliable and efficient approach for the separation and detection of morin from natural products and food samples.

Celastrol and Resveratrol Modulate SIRT Genes Expression and Exert Anticancer Activity in Colon Cancer Cells and Cancer Stem-like Cells

Helena Moreira et al.

Summary: The recovery rate in patients with metastatic colorectal cancer is low due to drug resistance and the presence of colorectal cancer stem cells (CSCs). Phytochemicals like -celastrol and resveratrol have shown promise in colon cancer therapy due to their low toxicity and ability to interact with various biological targets. This study investigates the anticancer mechanisms of these compounds against metastatic colon cancer cells and their capacity to eradicate CSCs.

Single-cell transcriptomic profiling unravels the adenoma-initiation role of protein tyrosine kinases during colorectal tumorigenesis

Xiaobo Zheng et al.

Summary: The study characterized cell types, transcriptional signatures, and differentially expressed genes at each stage of colorectal cancer development using single-cell RNA sequencing, identifying adenoma and carcinoma precursor cell populations and potential drivers of adenoma initiation. The overexpression of these drivers promoted colorectal epithelial cell proliferation and resulted in the formation of polyp-like buds. These findings enhance our understanding of colorectal cancer and may contribute to improved diagnosis and treatment.

Cancer Chemopreventive Activities of Silibinin on Colorectal Cancer through Regulation of E-Cadherin/β-Catenin Pathway

Saba Sameri et al.

Summary: The study demonstrated that Silibinin has various anti-tumor activities such as inducing apoptosis, suppressing migration, eliminating CSCs, and attenuating EMT-related markers in CRC cells. These effects are likely mediated through the blockage of the beta-catenin signaling pathway, a key component of the Wnt signaling pathway involved in CRC development.

Summary: The long-term success of standard anticancer monotherapeutic strategies has been limited by tolerability issues, treatment resistance, and cancer relapse. Combination therapy has shown promising results in strengthening the anti-tumor potential, reducing toxic effects, and overcoming resistance. Interest in combining natural products with conventional chemotherapeutics to improve treatment efficacy has been growing, with Thymoquinone (TQ) emerging as a promising candidate due to its low toxicity and anticancer efficacy.

Targeting a Lipid Desaturation Enzyme, SCD1, Selectively Eliminates Colon Cancer Stem Cells through the Suppression of Wnt and NOTCH Signaling

Yeongji Yu et al.

Summary: The study suggests that targeting stearoyl-CoA desaturase 1 (SCD1) may selectively eliminate cancer stem cells (CSCs) in colon cancer by inducing apoptosis and affecting the survival ability of CSCs through specific signaling pathways. The suppression effect of SCD1 inhibition on CSCs is primarily mediated by the downregulation of Notch and Wnt signaling genes, indicating the potential of targeting SCD1 in clinical settings for colon cancer treatment.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
