Relatlimab and Nivolumab versus Nivolumab in Untreated Advanced Melanoma

Hussein A. Tawbi et al.

Summary: LAG-3 and PD-1 are two distinct inhibitory immune checkpoints that contribute to T-cell exhaustion. The combination of relatlimab and nivolumab has been shown to be safe and effective in previously treated melanoma patients, and this study aimed to evaluate its safety and activity in previously untreated melanoma patients. The results demonstrated that the combination therapy provided a greater benefit in terms of progression-free survival compared to nivolumab alone, without any new safety signals.

Upregulation of VCAM-1 in lymphatic collectors supports dendritic cell entry and rapid migration to lymph nodes in inflammation

Jorge Arasa et al.

Summary: DC migration to dLNs is a slow process involving DCs entering lymphatic capillaries, crawling into lymphatic collectors, and rapidly transported to dLNs. Inflammation induces BM degradation and upregulation of VCAM-1 in collectors, facilitating DC entry into collectors in a CCR7- and beta 1 integrin-dependent manner. Loss of beta 1-integrins in DCs or VCAM-1 in collectors has the greatest impact on DC migration to dLNs at early time points.

MHC Class II Antigen Presentation by Lymphatic Endothelial Cells in Tumors Promotes Intratumoral Regulatory T cell-Suppressive Functions AC

Anastasia O. Gkountidi et al.

Summary: This study demonstrates that tumoral lymphatic endothelial cells (LEC) can promote tumor growth by upregulating MHC class II and antigen-processing machinery, leading to Treg expansion and suppression of effector T-cell infiltration. Lack of MHC class II expression in LECs impairs tumor growth, increases effector T-cell infiltration, and alters Treg-suppressive functions. Antigen presentation by LECs locally in the tumor microenvironment is crucial for promoting Treg-mediated immunosuppression.

Summary: Lymph node metastasis is a significant prognostic factor in various cancers, serving as a gateway for further dissemination. Prior to metastasis, the primary tumor reshapes the draining lymph node by secreting soluble factors or releasing extracellular vesicles, creating a pre-metastatic niche for the survival and growth of metastatic cells.
