Flagellar length control in multiflagellated eukaryotes: a case study with Giardia

S. Patra et al.

Summary: This article discusses the relationship between organism size and function, focusing on the control mechanism of flagellar length in eukaryotic cells. By analyzing a theoretical model and experimental data, the researchers successfully predicted the lengths of the four pairs of flagella in Giardia, an octo-flagellate.

Origins of eukaryotic excitability

Kirsty Y. Wan et al.

Summary: Eukaryotes have evolved radically new ways to sense and react to their environment, enabling more complex forms of cellular behavior. Key events and innovations during eukaryogenesis, including an expanded repertoire of ion channels and the relocation of chemiosmotic ATP synthesis to mitochondria, greatly amplified the degrees of freedom associated with cellular responses, allowing eukaryotes to outperform prokaryotes in terms of both speed and accuracy.

The many modes of flagellar and ciliary beating: Insights from a physical analysis

Charles B. Lindemann et al.

Summary: The article discusses the mechanism that allows the axoneme of eukaryotic cilia and flagella to produce both helical and planar beating, presenting two possibilities to explain the mechanism of planar beating and suggesting that the flagellum behaves as two ribbons of doublets interacting with a central partition. Additionally, the analysis examines the role of torsion in transitioning from a planar to a helical beating modality in long flagella. Finally, the article proposes tests to evaluate the theoretical and experimental aspects of their proposals.

Flagellar length control in monoflagellates by motorized transport: Growth kinetics and correlations of length fluctuations

Swayamshree Patra et al.

Summary: This study investigates how cells self-organize to control the lengths of their appendages, specifically focusing on the flagellum. By using a time of flight mechanism, a stochastic model was developed for two types of unicellular organisms, accurately predicting the temporal variation in flagella lengths. Furthermore, molecular communication within cells during the multiflagellated stage of the cell cycle was analyzed using Monte Carlo simulation.

Competitive binding of independent extension and retraction motors explains the quantitative dynamics of type IV pili

Matthias D. Koch et al.

Summary: Researchers labeled TFP in Pseudomonas aeruginosa and tracked extension and retraction cycles of individual filaments, finding that dynamics are stochastic and TFP can pause between extension or retraction events. They developed a biophysical model based on stochastic binding of extension and retraction motors, showing that the retraction motor dictates pilus production rate and is bound to pilus machines even in their unpiliated state. JSNameOur model quantitatively agrees with a variety of experiments, suggesting that the competition of stochastic motor binding to the pilus machine coordinates the repetitive cycle of pilus extension and retraction.

Analysis of biological noise in the flagellar length control system

David Bauer et al.

Summary: Cell-to-cell variation is mainly influenced by cell size, while within-cell variation is caused by dynamic fluctuations. Tubulin assembly is not directly related to intraflagellar transport, and length fluctuations reflect tubulin assembly and disassembly events involving large numbers of tubulin dimers. Length variation increases in long-flagella mutants, consistent with theoretical models for flagellar length regulation.

Robustness and the evolution of length control strategies in the T3SS and flagellar hook

Maulik K. Nariya et al.

Summary: The study focuses on a mathematical model of length control based on the ruler mechanism in bacterial cells, showing that the predictions of this model are consistent with experimental data. Compared to the substrate switching mechanism, the ruler mechanism allows for needles with larger average lengths without a concomitant increase in variance.
