Orally-active, clinically-translatable senolytics restore a-Klotho in mice and humans

Summary: This study demonstrates that targeting senescent cells can increase a-Klotho levels in mice and humans, suggesting a potential therapeutic avenue for enhancing a-Klotho. Removing senescent cells can restore a-Klotho expression, offering a translational feasible strategy for developing orally-active small molecule interventions. Urinary a-Klotho may also serve as a useful biomarker for monitoring senolytic clinical trials.

Drosophila fabp is required for light-dependent Rhodopsin-1 clearance and photoreceptor survival

Summary: This study highlights the role of Drosophila fatty acid binding protein (fabp) in regulating light-dependent Rhodopsin-1 (Rh1) proteostasis and photoreceptor survival. The induction of fabp in ninaE(G69D) mutant photoreceptors required vitamin A and its transporter genes, showing a link between nutrients and protein clearance. Loss of fabp resulted in accumulation of Rh1 proteins in cytoplasmic vesicles, leading to light-dependent retinal degeneration.

Green light extends Drosophila longevity

Summary: The effect of visible light on Drosophila melanogaster longevity is wavelength specific, with green light significantly extending lifespan and blue light dramatically reducing it. The beneficial effect of green light is not linked to oxidative stress, heat stress, or caloric restriction, and may be mediated by microbiota or photosensitive micronutrients in food medium. This suggests the potential of green light as a noninvasive therapy for aging-related diseases.
