Present and future of microglial pharmacology

Eva Simoncicova et al.

Summary: Microglia, as brain resident immune cells, play a crucial role in the development, activity, and plasticity of the central nervous system. They have been identified as potential targets for new neurotherapies due to their involvement in various neurological pathologies. Current research focuses on understanding the complex heterogeneity of microglia and developing more targeted treatments. Although the current therapies have limitations, clinical investigations are underway, and the future of microglial therapeutic research looks promising.

Microglial large extracellular vesicles propagate early synaptic dysfunction in Alzheimer's disease

Martina Gabrielli et al.

Summary: The study reveals that microglial extracellular vesicles carrying amyloid-beta can move along axons and spread synaptic dysfunction. Inhibiting the motion of these vesicles restricts the spread of dysfunction, suggesting a potential strategy to delay Alzheimer's disease progression.

Soluble TREM2 in CSF and its association with other biomarkers and cognition in autosomal-dominant Alzheimer's disease: a longitudinal observational study

Estrella Morenas-Rodriguez et al.

Summary: This study aimed to investigate the associations between soluble TREM2, a biomarker of TREM2 signaling, and amyloid beta deposition, tau-related pathology, neuroimaging markers, and cognitive decline in the progression of Alzheimer's disease. The results demonstrated that increased soluble TREM2 was associated with amyloid beta deposition, tau pathology, cortical shrinkage, and cognitive decline.

BACE-1 inhibition facilitates the transition from homeostatic microglia to DAM-1

Neeraj Singh et al.

Summary: BACE-1 plays a role in regulating the transition of microglia in Alzheimer's disease. Deficiency of BACE-1 leads to increased expression of certain transcription factors involved in the transition from homeostatic to highly phagocytic microglia. Similar transition-state microglia in AD brains are associated with decreased levels of BACE-1 expression.

Targeting microglial autophagic degradation in NLRP3 inflammasome-mediated neurodegenerative diseases

An-Guo Wu et al.

Summary: Neuroinflammation is considered detrimental in neurodegenerative diseases, with NLRP3 inflammasome playing a crucial role in exacerbating pathology and disease progression. Microglial autophagy is important in maintaining brain homeostasis and negatively regulating NLRP3 inflammasome-mediated neuroinflammation. Understanding sex differences in NLRP3 inflammasome-mediated neurodegenerative diseases will aid in developing targeted therapies and improving diagnostic and prognostic abilities.

The role of innate immune genes in Alzheimer's disease

Ana Griciuc et al.

Summary: Recent studies have highlighted the crucial role of the innate immune system and neuroinflammation in the pathogenesis of Alzheimer's disease, with a focus on microglial receptors CD33 and TREM2. Therapeutic approaches targeting CD33 and TREM2, such as gene therapy, small molecules, and immunotherapy, have shown promise in modulating neuroinflammation and potentially reducing neurodegeneration in Alzheimer's disease. Antibodies against CD33 and TREM2 have entered clinical trials and may provide a new avenue for treating Alzheimer's disease by targeting neuroinflammation.

Targeting Microglia-Synapse Interactions in Alzheimer's Disease

Gaia Piccioni et al.

Summary: This review focuses on the emerging roles of microglia in the brain, particularly in synaptic plasticity in health and disease. Evidence suggests that maintaining the ramified state of microglia is crucial for normal synaptic plasticity and cognitive function. Inflammatory signals associated with diseases like Alzheimer's can alter the morphology of microglia and lead to synaptic loss and cognitive impairment, highlighting potential targets for disease-modifying therapy.

Therapeutic Targeting of the Complement System: From Rare Diseases to Pandemics

Peter Garred et al.

Summary: The complement system is the host's defense friend by protecting it from invading pathogens, promoting tissue repair, and maintaining homeostasis. Complement is a double-edged sword, since when dysregulated or overactivated it becomes the host's enemy, leading to tissue damage, organ failure, and, in worst case, death. A number of acute and chronic diseases are candidates for pharmacological treatment to avoid complement-dependent damage, ranging from the well established treatment for rare diseases to possible future treatment of large patient groups like the pandemic coronavirus disease 2019.
