Manufacturing Process Development for Belzutifan, Part 6: Ensuring Scalability for a Deoxyfluorination Reaction

Summary: This article presents the development of a regio- and stereoselective deoxyfluorination process for the manufacture of belzutifan (MK-6482). Through multiple studies, including mechanistic, range-finding, and mixing studies, strategies were identified to overcome the challenges posed by specific reaction and engineering sensitivities. The discovery of a second phase of perfluoro-1-butanesulfonyl fluoride in the reaction at cryogenic temperature during mixing studies led to the implementation of controls to minimize the formation of side products. These improvements resulted in a successful execution of the process on pilot and commercial scales.
