Electrochemical utilization of methanol and methanol-d4 as a C1 source to access (deuterated) 2,3-dihydroquinazolin-4(1H)-one

Summary: An electrocatalytic protocol for the synthesis of 2,3-dihydroquinazolin-4(1H)-one has been disclosed. Methanol is activated and used as the C1 source to cyclize with 2-aminobenzamides, resulting in a convenient cyclization reaction without the need for homogeneous metal catalysts, external oxidants, or bases. A wide variety of N,N-disubstituted 2,3-dihydroquinazolin-4(1H)-ones can be obtained via this approach.

Photoinduced Transition-Metal-Free Chan-Evans-Lam-Type Coupling: Dual Photoexcitation Mode with Halide Anion Effect

Summary: In this study, we report a photoinduced transition-metal-free C(aryl)-N bond formation method using 2,4,6-tri(aryl)boroxines or arylboronic acids as an aryl source and 1,4,2-dioxazol-5-ones as an amide coupling partner. The presence of chloride anion, either generated in situ by the photodissociation of chlorinated solvent molecules or added separately, plays a critical cooperative role in facilitating the synthesis of a wide range of synthetically versatile N-arylamides under mild photo conditions. Mechanistic investigations suggest the involvement of two distinctive photoexcitations in the process, enabling the activation of dioxazolones and the migration of aryl groups.

Summary: By employing N-hydroxypyridin-2-ones as the oxygen and nitrogen sources, arylboronic acids were difunctionalized successfully to prepare various N-(2-hydroxyaryl)pyridin-2-ones in good yields through a copper(II)-catalyzed Chan-Lam reaction and subsequent BF3 promoted selective 1,3-rearrangement. Mechanistic studies revealed that the selectivity of the 1,3-rearrangement is controlled by the formation of the key aryloxypyridinium salt. The obtained products can be easily converted to various useful pyridin-2-one scaffolds.

Synthesis of quinazolin-4(3H)-ones via electrochemical decarboxylative cyclization of α-keto acids with 2-aminobenzamides

Summary: An environmentally friendly electrochemical method was disclosed for the synthesis of quinazolin-4(3H)-one derivatives from readily available alpha-keto acids and 2-aminobenzamides. The decarboxylative cyclization process proceeds conveniently without the need for homogeneous metal catalysts, bases, or external oxidants. The protocol also features CO2 by-products, mild reaction conditions, and a wide variety of substrate scope, including an array of 2,3-disubstituted quinazolinone products.

Mediator-Enabled Electrocatalysis with Ligandless Copper for Anaerobic Chan-Lam Coupling Reactions

Summary: This study reports the use of substoichiometric quantities of redox mediators to address limitations to Cu-catalyzed electrosynthesis. By applying this strategy to Chan-Lam coupling of aryl-, heteroaryl-, and alkylamines with arylboronic acids without the need for chemical oxidants, couplings under these electrochemical conditions occur with higher yields and shorter reaction times compared to conventional reactions. The redox mediators serve multiple roles in rapidly oxidizing low-valent Cu intermediates, regenerating the catalyst, revealing the active Pt surface for proton reduction, and providing anodic overcharge protection to prevent substrate oxidation.
