Organocatalysis: A recent development on stereoselective synthesis of o-glycosides

Ram Naresh Yadav et al.

Summary: Organocatalysis is a method of using low-molecular-weight organic molecules to accelerate stereoselective chemical transformations. It has recently gained attention in synthetic and medicinal chemistry for its potential in asymmetric synthesis of important compounds. Organocatalysts have advantages over metal-based catalysts, such as better tolerance to air and moisture, easy availability, easy handling, non-toxicity, and high enantiomeric purity. This perspective focuses on the application of asymmetric organocatalysis in the synthesis of biologically significant O-glycosides.

Recent Chemical and Chemoenzymatic Strategies to Complex-Type N-Glycans

Xiaoya Zhao et al.

Summary: Glycosylation is a major form of protein post-translational modification, and N-glycans play a crucial role in intercellular interaction and immune function. Efficient methods to provide homogeneous complex-type N-glycans have been urgently needed for analytical and medicinal purposes. Significant progress has been made in both chemical and chemoenzymatic strategies for the synthesis of N-glycans.

Recent advances in stereoselective 1,2-cis-O-glycosylations

Akihiro Ishiwata et al.

Summary: 1,2-cis-O-glycosylation is essential in the assembly of bioactive glycans with various functions. However, controlling the stereoselectivity of glycosylation and specifically forming 1,2-cis glycosides is challenging. Recent advances in glycosyl donor modification, reaction conditions, and activation methods have been made in the development of 1,2-cis selective glycosylations.

Zinc(II) Iodide-Directed β-Mannosylation: Reaction Selectivity, Mode, and Application

Xuemei Zhong et al.

Summary: The study presents a strategy for direct mannosylation mediated by ZnI2, allowing for highly stereoselective construction of 1,2-cis-beta linkages, which can be applied to various alcohol acceptors for beta-mannosylation and modular synthesis of trisaccharides for N-glycan synthesis. Theoretical investigations using density functional theory calculations elucidated the mechanistic details of this beta-selective mannosylation.

O-Glycosyl Trichloroacetimidates as Glycosyl Donors and Platinum(IV) Chloride as a Dual Catalyst Permitting Stereo- and Regioselective Glycosidations

Tong Li et al.

Summary: Stereo- and regioselective glycosidation remains a significant challenge in carbohydrate chemistry. Acid-base catalysis, particularly in O-glycosyl trichloroacetimidate activation, offers an alternative pathway with increased proton acidity and oxygen nucleophilicity for selective glycosidation. Platinum(IV) chloride serves as an ideal catalyst due to its weak Lewis acid properties and bidentate ligation capacity, enabling predominantly beta-configuration glycosidation products and regioselectivity in various carbohydrate acceptor moieties.
