Dose mediates the protracted effects of adolescent THC exposure on reward and stress reactivity in males relevant to perturbation of the basolateral amygdala transcriptome

Jacqueline-Marie N. Ferland et al.

Summary: Contrary to the belief that cannabis is harmless, exposure to it during adolescence increases the risk of developing psychopathologies in adulthood. This study found that repeated exposure to low and high doses of THC during adolescence had different effects on behavior in adulthood. The study also revealed disturbances in gene networks related to reward processing and stress, as well as dysregulation of astrocytes.

The Cannabinoid Receptor Agonist, WIN, Suppresses the Activation of Proinflammatory Genes Induced by Interleukin 1 Beta in Human Astrocytes

Jerel Adam Fields et al.

Summary: The study demonstrated that WIN can effectively inhibit the inflammatory response in reactive astrocytes by modulating gene expression patterns, particularly those related to neuroprotective transcription factors. The anti-inflammatory properties of WIN are associated with the regulation of kinase pathways and are independent of CB1, suggesting alternative receptors mediate its effects. This provides mechanistic insights into the anti-inflammatory activity of WIN and highlights astrocytes as a potential therapeutic target for ameliorating neuroinflammation.

Modulating the Neuromodulators: Dopamine, Serotonin, and the Endocannabinoid System

Kate Z. Peters et al.

Summary: Dopamine, serotonin, and endocannabinoids play crucial roles in motivated behavior, including reward processing and reinforcement learning. The interactions between these neuromodulatory systems are complex, and recent evidence suggests potential new relationships between them, challenging longstanding views. Advanced technologies for precise identification and control of genetically targeted neuronal populations are crucial for understanding the intricate relationship between these essential systems and their impact on motivated behavior.

Repeated exposure to JWH-018 induces adaptive changes in the mesolimbic and mesocortical dopaminergic pathways, glial cells alterations, and behavioural correlates

Nicholas Pintori et al.

Summary: Repeated exposure to JWH-018 induced anxious and aversive behaviors, decreased dopamine neurons in the VTA, and altered dopaminergic transmission. The drug also affected dopamine sensitivity in the NAc shell and core, while causing neuroinflammatory responses in specific brain areas. This study suggests that recurring use of Spice/K2 drugs may have detrimental effects on behavior and dopamine regulation.

Summary: The study found that parvalbumin (PV)-expressing neurons play a key role in the pathology of autism spectrum disorders (ASD). Deficiency in the Kv3.1 channel can induce social deficits, hyperactivity, and stereotypic behaviors independent of changes in GAD67 levels.

Rapamycin prevents the long-term impairing effects of adolescence Δ-9-tetrahydrocannabinol on memory and plasticity in male rats

Talia Sabran-Cohen et al.

Summary: The study found that adolescent exposure to THC can have long-term effects on object recognition short-term memory and synaptic plasticity, but late-adolescent exposure did not show the same effects. Rapamycin was able to rescue these persistent effects of THC during adolescence.

Functional validation of a finding from a mouse genome-wide association study shows that Azi2 influences the acute locomotor simulant response to methamphetamine

Xinzhu Zhou et al.

Summary: The study aimed to investigate the role of Azi2 in methamphetamine sensitivity by creating mutant Azi2 alleles and analyzing their effects on the locomotor response to methamphetamine. While the mutant allele increased sensitivity, contrary to GWAS results, lower Azi2 levels did not necessarily decrease the response. Despite no direct correlation between Azi2 and Slc6a3, the Azi2 mutation altered the expression of important genes and pathways related to methamphetamine sensitivity.
