Summary: The loss of dopamine-producing neurons in the substantia nigra pars compacta (SN) is the basis of the core motor symptoms of Parkinson's disease (PD). Current treatments do not exist to prevent or slow the neurodegeneration of these neurons, and factors like disrupted Ca2+ homeostasis and mitochondrial dysfunction may contribute to their vulnerability. Various types of Ca2+ channels, such as L-type voltage-gated Ca2+ channels (LTCCs) and R- and T-type Ca2+ channels (RTCCs and TTCCs), are being explored as potential drug targets for PD therapy, but recent trials have shown mixed results in slowing disease progression.
