The potential of microbiome replacement therapies for Clostridium difficile infection

Anthony M. Buckley et al.

Summary: Recent studies have shown that using microbiome therapy to restore bile homeostasis and reduce the bioavailability of preferential nutrients can effectively target the germination ability of C. difficile spores and the growth rate of vegetative cells in treating C. difficile infection. Several biotechnology companies are developing microbiome therapies for CDI treatment, with clinical trials in progress.

Clostridioides difficile toxins: mechanisms of action and antitoxin therapeutics

Shannon L. Kordus et al.

Summary: Clostridioides difficile produces up to three different toxins, which play a key role in the pathogenesis of colon infection. In this Review, Kordus, Thomas, and Lacy discuss the structure and function of these toxins and how this information guides new therapeutic approaches. Understanding the mechanisms of host-toxin interactions provides a foundation for developing novel strategies for the treatment and prevention of C. difficile infection.

TFPI is a colonic crypt receptor for TcdB from hypervirulent clade 2 C. difficile

Jianhua Luo et al.

Summary: The study identified tissue factor pathway inhibitor (TFPI) as the receptor for TcdB4, a dominant virulence factor of hypervirulent clade 2 Clostridioides difficile. TFPI is highly expressed in the intestinal glands and protects the colonic epithelium from TcdB2/4. These findings reveal new mechanisms for CDI pathogenesis.

Low-density lipoprotein receptor-related protein 1 is a CROPs-associated receptor for Clostridioides difficile toxin B

Shengjie Guo et al.

Summary: Our study identified LDL receptor-related protein-1 (LRP1) as a novel receptor for TcdB, providing evidence for a new mechanism of TcdB entry. Knockout of LRP1 in cells increased resistance to TcdB, while overexpression of LRP1 sensitized cells to low concentrations of TcdB, suggesting potential therapeutic targets for treating C. diff.

Host Immune Responses to Clostridioides difficile: Toxins and Beyond

Britt Nibbering et al.

Summary: Clostridioides difficile, a bacterium often resistant to antibiotics, is a leading cause of nosocomial infectious diarrhea worldwide due to toxin production. Studies suggest that immune responses to non-toxin proteins may play an important role in protection against CDI in humans.

Large Clostridial Toxins: Mechanisms and Roles in Disease

Kathleen E. Orrell et al.

Summary: Large clostridial toxins (LCTs) are a family of bacterial exotoxins that infiltrate and destroy target cells, implicated as the primary virulence factors in various infections. Understanding the mechanism of LCT intoxication through recent structural work and toxin subtyping analysis is crucial for elucidating their role in infection and disease. Research on genomic discovery and characterization of LCT homologues provides valuable insights into this enigmatic family of toxins, offering perspectives on emerging questions and future research directions.
